Embodiment Construction

[0017] The present invention will be described in further detail below in conjunction with the embodiments and accompanying drawings.

[0018] see Figure 6 , a fast and convenient method for making slices of dense filamentous bacteria, first use a scalpel to draw a straight line perpendicular to the surface of the medium near the edge of the dense filamentous bacteria colony in the petri dish, and then along the outside of the edge of the filamentous fungal colony Draw an arc, and cut off a small piece of 1-2mm wide culture together with the agar; then gently pick it up with a scalpel, and flatten the cut agar surface on the glass slide, so that the cut surface of the knife is close to the slide. Slides; finally, microscopic examination was performed from low magnification to high magnification.

Abstract

The invention provides a rapid and convenient sheet preparing method of compact filamentous bacteria and relates to the technical field of filamentous bacteria sheet preparation. The method comprises the following steps: scribing one line perpendicular to the surface of a culture medium and near the edge of bacterial colony of the filamentous bacteria in a culture dish by a surgical knife blade, scribing an arc line along the outer side of the edge of bacterial colony of the filamentous bacteria, and cutting of a small culture object with width being 1 to 2 mm together with agar; stirring up the culture object slightly with the surgical knife, and flatly paving the cut agar surface on a glass slide to enable the knife cutting surface to abut against the glass slide; performing microscopic examination from low power to high power. The sheet preparing method provided by the invention is rapid and convenient; long-time cultivation is not needed; the original mycelial morphology is completely not damaged, the original and natural growth state of the filamentous bacteria is maintained, the method is particularly suitable for typical for observation of substrate mycelium such as spore hypha, aerial hyphae, rhizoid and stolon of compact filamentous bacteria (comprising undeveloped filamentous bacteria such as actinomycetes and aerial hyphae), and the morphological characteristics of different growth stages can be observed simultaneously.

Description

technical field [0001] The invention relates to the technical field of filamentous fungus sheet production, in particular to a fast and convenient method for preparing dense filamentous fungus sheets. Background technique [0002] In the microbiology experiment class, to observe the morphology of filamentous bacteria (mold and actinomycetes), first of all, it is necessary to use a glass slide to prepare a specimen piece, and then use a microscope to examine it. In routine slide production, three methods are usually used: direct slide slide production, slide wet chamber culture slide production, and scotch tape paste slide production. These three methods either easily destroy the original mycelium morphology and structure, or the preparation method is cumbersome and time-consuming. [0003] see figure 1 As shown, in the petri dish, the fungal filamentous colony grows on the agar solid medium. When making slices, the hyphae are generally picked directly with an inoculation l...
