Embodiment Construction

[0031] A method for extracting organic phosphorus from lake algae, comprising the following steps:

[0032] (1) Sample collection: collect floating algae on the surface of eutrophic lake water, put them in a sealed bag for refrigerated storage, transport them back to the laboratory for freeze-drying, grind them thoroughly after freeze-drying, and pass through a 2mm sieve to obtain algae powder samples, which are frozen at -20°C Save for later use;

[0033] (2) Determination of the basic composition of phosphorus: total phosphorus TP, inorganic phosphorus P i and P o the determination of

[0034] Accurately weigh two samples of 0.5g algae powder, each with 3 parallel samples, one was calcined in a muffle furnace at 450°C for 3 hours, and transferred to a centrifuge tube after cooling down to room temperature, and 20 mL of 3.5 mol / L HCl was added to shake for 16 hours. After centrifugation, the supernatant was filtered with a 0.45 μm filter membrane to obtain the extract, dig...

Abstract

An extraction and composition analysis method for lake alga organophosphorus is disclosed. The method includes steps of sample collection, measurement of phosphorous basic composition, organophosphorus (Po) extraction, sample testing pretreatment, liquid phosphorus nuclear magnetic resonance (<31>P-NMR) analysis and calculation. A mixture of 0.5 mol / L of NaOH and 25 mmol / L of EDTA is adopted as an extractant, and the solid liquid ratio of an alga powder sample to the extractant is 1:60 in the method to obtain an optimum Po extraction effect. During <31>P-NMR analysis testing, delay time (D1) is set to be 5 s and scanning analysis time is 15 h so that a good spectrum can be obtained. The average extraction rates of total phosphorous (TP) and the Po in the method are respectively 95.0% and 94.9% or above. The method can accurately detect specific varieties and contents of the Po in the sample, and can truly and accurately reflect composition characters of the Po in the lake algae. The method provides important scientific bases for researching contributions of lake algae in an algal bloom process, for researching lake endogenous phosphorus circulation processes and for government decision-making.

Description

technical field [0001] The invention relates to a method for extracting and analyzing the composition of organic phosphorus, in particular to a method for extracting and analyzing the composition of organic phosphorus in lake algae. Background technique [0002] Phosphorus is one of the key elements in the metabolism of freshwater lake ecosystems. Excessive phosphorus will lead to the growth and reproduction of algae, causing deterioration of water quality and endangering the safety of ecosystems. Exogenous input and endogenous circulation are the main factors leading to the substantial increase of phosphorus in lake water. In recent years, the input of exogenous phosphorus, including domestic sewage, industrial and agricultural production, and soil runoff, has been controlled to a certain extent, and the release of endogenous phosphorus in lakes has become a key factor causing further deterioration of lake water quality. Sediment release is one of the important sources of ...
