Problems solved by technology

However, there are still some problems in its application, such as: most light-sensitive hydrogel materials are chemically synthesized, and there are problems with cell compatibility, and due to the lack of cell adhesion groups, cells cannot attach and grow, which greatly affects cell function
In addition, the crosslinking agents currently used for photocrosslinking are only sensitive to ultraviolet light, and the use of ultraviolet light has the potential to have a mutation effect on the cells contained in the gel and the surrounding host tissue, which cannot be applied clinically.

[0064] Preparation Example 1: Preparation of Photocrosslinked Gelatin

[0065] 1) Dissolve 15g of collagen gelatin (Gelatin) powder purchased from Sigma-Aldrich (product number: G9391) in 500ml of water, and place it in a shaker at 37°C until the gelatin is completely dissolved to form a 3% (w / v) transparent solution;

[0066] 2) Add 6ml, 12ml and 30ml of methacrylic anhydride (>94%, Sigma-Aldrich, 276685) to the resulting solution, respectively, so that the molar ratio of gelatin to methacrylic anhydride is 1:10, 1:20 and 1, respectively :50, vigorously shaken and mixed, placed in a shaker at 37°C overnight;

[0067] 3) Take out the reacted mixture, pour it into a dialysis bag (molecular weight cut-off 3.5k), and dialyze in water (4L) at room temperature for 2 days, changing the water at least 10 times during the period;

[0068] 4) Take out the purified methacrylated gelatin, use Benchtop FreeZone4.5L (Labconco) to lyophilize into a sponge-like solid, and place it in a de...

[0069] Preparation Example 2: Preparation of Photocrosslinked Hyaluronic Acid

[0070]Dissolve 5g of hyaluronic acid (MW600,000-850,000, HA700K-1, Lifecore) in 500ml of water and add to 9ml, 18ml and 36ml of methacrylic anhydride respectively, so that the molar ratio of hyaluronic acid and methacrylic anhydride They are 2:1, 1:1 and 1:2 respectively. The reaction conditions and purification steps were the same as those in Preparation Example 1, and the molecular weight cut-off of the dialysis bag was 3.5k.

[0071] Preparation Example 3: Preparation of Photocrosslinked Chitosan

[0072] Dissolve 7g of chitosan (MW140,000~220,000, 740179, Sigma) in 500ml of 2% (v / v) acetic acid (320099, Sigma) solution, add to 7.5ml, 15ml and 30ml of methacrylic anhydride, respectively , so that the molar ratios of chitosan and methacrylic anhydride were 2:1, 1:1 and 1:2, respectively. The reaction conditions and purification steps were the same as those in Preparation Example 1, and the molecular weight cut-off of the dialysis bag was 3.5k.

Abstract

The invention provides a photo-crosslinking material which is composed of a methacryliated material containing -OH and -NH2 groups. The photo-crosslinking material is prepared by the following method: 1) the material containing -OH and -NH2 groups is dissolved; and 2) a solution obtained and methacrylic anhydride react for at least 16 h. The invention also provides hydrogel formed by the photo-crosslinking material. The hydrogel is prepared by the following method: 1) a light initiator is dissolved in a transparent solution of the photo-crosslinking material when protected from light; and (2) a solution obtained is illuminated. The hydrogel can be used as a repairing material of wounded tissues.

Description

technical field [0001] The invention relates to a photocrosslinking material, a hydrogel formed by the photocrosslinking material, a preparation method and an application thereof. Background technique [0002] Tissue defects in the human body caused by trauma and disease require biomaterials to provide support for tissue growth, and these biomaterials can also serve as transport carriers for cells and regenerative factors. However, the shape of wounds in the body is often irregular. This problem can be partially solved by pruning the material, but there are still parts where the host tissue does not match the graft material, which greatly reduces the integration of the graft material with the host tissue and the subsequent repair effect. [0003] Solid Freeform Fabrication (Solid Freeform Fabrication) technology can effectively solve this problem. The materials used in this kind of technology exist in the state of powder or solution at first, so that it can perfectly and se...
