Embodiment 1

[0026] (1) Pretreatment of carrier materials

[0027]Corn stalks, wheat stalks and sycamore leaves collected in farmland and the field were dried, crushed, and passed through a 24-mesh sieve; then mixed with commercially available wheat bran and self-made 24-mesh walnut shell activated carbon. The proportion of the carrier material is 50wt% wheat straw, 20wt% corn straw, 5wt% wheat bran, 10wt% sycamore leaves, and 12.5wt% walnut shell activated carbon. Soak the mixed carrier in distilled water for 12-24 hours, adjust the pH to 7.0-7.5, filter out the floating impurities and supernatant in the upper layer, bake the filtered mixed carrier at 50°C for 24 hours, and sterilize it with high-pressure steam at 121°C for 20 minutes, and set aside.

[0028] Walnut shell activated carbon uses ZnCl 2 The activation method is prepared by weighing a certain amount of walnut shells that have been crushed through a 24-mesh sieve and immersed in 50wt% ZnCl 2 In the solution, the ratio of sol...

Embodiment 2

[0037] The difference from Example 1 is that the ratio of the carrier material is 37.5wt% of wheat straw, 20wt% of corn straw, 10wt% of wheat bran, 12.5wt% of sycamore leaves, and 20wt% of walnut shell activated carbon.

[0038] After repairing for 40 days under the same conditions, the removal rate of petroleum pollutants can reach 62wt%, and the removal rate of saturated hydrocarbons can reach 72.5wt%.

Embodiment 3

[0040] One of the differences from Example 1 is that the ratio of the carrier material is 25wt% of wheat straw, 30wt% of corn straw, 15wt% of wheat bran, 15wt% of sycamore leaves, and 15wt% of walnut shell activated carbon. The second difference is that the amount of immobilized petroleum-degrading bacteria sprayed into the finished product is 1kg / m 2 .

[0041] Other conditions are the same, the repair time is 40 days, the removal rate of petroleum pollutants can reach 58.5wt%, and the removal rate of saturated hydrocarbons can reach 70.3wt%.

Abstract

The invention discloses an immobilized method and application of petroleum degrading bacteria. The method includes the following steps: mixing and pretreating carrier materials of wheat straw, corn straw, wheat bran, leaves and walnut shell activated carbon; inoculating an activated petroleum degrading bacterium seed liquid, and carrying out adsorption culture; and filtering out an upper bacterial suspension under aseptic conditions, rinsing the carrier materials with sterile water, transferring the carrier materials adsorbed with bacteria to a new petroleum degrading bacterium seed liquid, repeating several times to reach saturation of the bacteria adsorbed by the carrier materials, so as to obtain the immobilized particles. The invention uses the natural organic materials and natural inorganic materials as carriers to overcome the defects of the individual usage of inorganic carrier or organic carrier; adsorption method is employed to realize immobilization of petroleum degrading bacteria; and the prepared immobilized petroleum degrading bacteria can be applied to in-situ remediation of petroleum contaminated soil to reach good effect, and is an environmentally friendly product which can be popularized and applied.

Description

technical field [0001] The invention relates to microorganism immobilization technology, in particular to an immobilization method and application of petroleum-degrading bacteria used for repairing petroleum-contaminated soil. Background technique [0002] During the process of oil extraction, refining, transportation and use, various reasons will cause different degrees of leakage, which will cause oil pollution to the soil. Petroleum-contaminated soil is one of the important pollutions of petroleum and petrochemical enterprises, and the petroleum pollutants in it have a high poisonous effect on human body and environment. The treatment methods of petroleum-contaminated soil at home and abroad mainly include incineration, landfill, stacking and bioremediation. [0003] The core of microbial remediation technology is the degradation of petroleum pollutants by highly efficient degrading bacteria. The degradation process makes the composition of pollutants tend to be more com...
