Embodiment 1

[0052] In this example, three sets of primer pairs were designed and synthesized, and on the basis of the primer pairs, three (TA) assays for detecting the ESR1 gene associated with susceptibility to liver cancer were designed. n , T29C and Pvu II loci genotype and by (TA) n , T29C, Pvu II and Xba I sites composed of haplotype method.

[0053] 1. Genomic DNA extraction

[0054] Genomic DNA of peripheral blood leukocytes was extracted by Miller's modified salting-out method, which is conventional in the field.

[0055] 1) Take sodium citrate or EDTA-Na 2 5ml of anticoagulated whole blood (try not to use heparin for anticoagulation), put in a 50ml capped centrifuge tube;

[0056] 2) Add 3-5 times the volume of cold distilled water, invert and mix repeatedly, place in ice for 5 minutes, and centrifuge at 2000rpm at 4°C for 20 minutes;

[0057] 3) Slowly pour the supernatant, add pre-cooled 0.1% Triton-X100 (same volume as above) to the precipitate, gently mix the precipitate,...

Embodiment 2

[0074] This embodiment uses the method that embodiment 1 sets up, to 248 liver cancer patients and 239 matched HBV carrier control populations without liver cancer (TA) n , T29C and Pvu II locus genotype and by (TA) n , T29C, Pvu II and Xba I loci composed of haplotypes were analyzed.

[0075] 1. Test object:

[0076] The study population consisted of 487 individuals. 248 patients with liver cancer, including 218 males and 30 females, aged 21-77 years (average 46.9 years old), all of them were from Guangxi, from Guangxi Zhuang Autonomous Region Cancer Hospital (Nanning, China), diagnosed by clinical and pathological sections HCC, without treatment; 239 healthy controls, including 199 males and 40 females, aged 18-80 years (average 47.2 years old), all from Guangxi, from the same area and the same time period as the HCC cases Those who were randomly screened out during early cancer screening, had no family history of liver cancer, and had no blood relationship with the indiv...

Abstract

This invention proposes a detection method of haplotype comprised of polymophic loci (rs3138774) in (TA)n, rs2077647 in T29C, rs2234693 in Pvu II and (TA)n, T29C, Pvu II and Xba I in susceptibility gene ESR1 relates to liver cancer risk. The methods are PCR-RFLP (polymerase chain reaction-restriction fragment length polymorphism), fragment analysis and direct sequencing PCR products. The haplotype is constructed by PHASE program. The invention also relates to the kits detecting susceptibility polymophic locis and using methods. In addition, site(TA)n (rs3138774), T29C(rs2077647) , Pvu II(rs2234693) and the haplotype comprising of (TA)n, T29C, Pvu II and Xba I were related to liver cancer risk. ESR1 is determined as a new susceptibility gene of liver cancer, which provides new genetic consultation content for the liver cancer prevention and control.

Description

technical field [0001] The present invention relates to the individual susceptibility and genetic detection method in crowd prevention of liver cancer. Specifically, the present invention relates to a genotype detection method for the polymorphism of the susceptibility gene ESR1 associated with the risk of liver cancer. The present invention further relates to a detection kit for the above-mentioned susceptibility gene ESR1 polymorphism genotype, and the application of the kit. In addition, the present invention also relates to the application of the above detection method and susceptibility gene in the prevention and control of liver cancer and genetic counseling. Background technique [0002] Primary hepatocellular carcinoma (referred to as liver cancer; Hepatocellular Carcinoma, HCC) is a common malignant tumor that seriously threatens human life and health. Epidemiological data show that liver cancer accounts for about 5% of new malignant tumor patients in the world ev...
