Problems solved by technology

Although this method can obtain water-soluble isoflavones, its concentration is low; and the whole method only considers isoflavones, which makes insufficient use of other ingredients in soybeans, and the cost is relatively high

Embodiment 1

[0020] Soybean isoflavone extract 400g and corn starch 1200g were dissolved in 60°C pH6.550mM 10L NaOC 2 h 5 -C 2 h 5 OOH buffer solution was incubated for 10 min, then cycloglucanotransferase (2 U / mL) was added, and reacted under the above conditions for 5 hr. After the reaction is finished, cool the reaction solution to room temperature, then centrifuge the reaction solution at 4000rpm / min for 10min, and the obtained supernatant flows through a 2.5×5cm AmberliteXAD-16 macroporous resin column, and washes with 2 times the volume of water Remove the soluble sugar compound and buffer, then elute the isoflavones adsorbed in the resin column with 2 times the volume of ethanol, and finally remove the ethanol by concentrating under reduced pressure to obtain a polysaccharide-based soybean isoflavone mixture.

Embodiment 2

[0021] Example 2. Dissolving 100 g of polysaccharide-based soybean isoflavones, 20 g of soybean oligosaccharides, and 10 g of calcium lactate in 5 L of pure water for medicine was prepared into calcium-added oral liquid of soybean isoflavones after sterilization.

Embodiment 3

[0022] Example 3. Dissolving 100 g of sterilized polysaccharide-based soybean isoflavones and 30 g of paclitaxel in 5 L of pure water for medicine, and then filtering through a 0.22 μm membrane to prepare soybean isoflavone paclitaxel injection. Before use, dilute 5 mL of this product into 0.9% sodium chloride injection or 500 mL of 5% glucose injection, use non-polyvinyl chloride infusion bottle and infusion tube, and filter through the connected 0.22 μm pore size filter , made into intravenous infusion.

Abstract

This invention belonging to chemical field is about preparation of high water-solubility soybean isoflavone and its derivative. By using cyclo-glucosyl transferase as enzyme and the highly dense starch as glycosyl donor, we convert natural soybean isoflavone to polysaccharide-isoflavone mostly complexed with cyclo-glucosyl. This invention is advantageous in high water-solubility compared with the natural ones. At the indoor temperature, its dissolvability is 95.2mM, 222 times higher than the natural soybean isoflavones. In the procedure, the aglycone structure hasn't been changed to remain natural activity in maximum. After filter and sterilization, the products could be injection, oral liquid and powder injection with cryodesiccation. It could be used in tumor (especially mammary cancer and prostate cancer), hectic fever, osteoporosis, hyperlipoidemia and so on. It overcomes the previous soybean isoflavone which could only be oral and which is low in biological effectiveness and therapeutic efficacy.

Description

technical field [0001] The invention belongs to the technical field of chemistry, and in particular relates to synthesizing required compounds by an enzymatic method. Background technique [0002] Synthetic estrogen used in the treatment of women's menopause syndrome can sometimes cause other diseases. In view of the fact that soy isoflavones have very good effects in preventing and reducing the incidence of cancer, increasing bone density, relieving hot flashes, and increasing the body's immunity, so it has become a commonly used ingredient in hormone replacement therapy (HRT) and is used for Oral drugs. [0003] However, many recent studies have confirmed that oral administration of soy isoflavones has low bioavailability (bioavailability), and its absorption is carried out in the small intestine (Turner NJ, Thomson BM, Shaw IC.Bioactive isoflavones in functional foods: the importance of gutmicroflora on bioavailability. Nutr Rev. 2003 Jun;61(6Pt 1):204-13.). Natural gl...
