Embodiment 1

[0027] Wash and dry 500 g of the crude drug Scrophulariaceae Scrophulariae, crush, soak in 5 liters of deionized water at room temperature for 24 hours, and filter. Continue soaking the residue in 2.5 liters of deionized water for 12 hours, filter, combine the filtrates, and concentrate under reduced pressure at 60°C to 1-2 liters. Then precipitate with 95% acetone, the final concentration of acetone is 80%, and precipitate for 24 hours. Centrifuge, dissolve the precipitate with 1.25 liters of water, centrifuge, and dialyze with deionized water for 48 to 72 hours. Concentrate under reduced pressure at 60°C and freeze-dry to obtain 30 g of total sugar from Scrophulariaceae. Add 2 liters of water to dissolve, add activated carbon to treat, remove twice, then concentrate under reduced pressure, centrifuge, and freeze-dry to obtain 15 g of total sugar of Scrophulariaceae with a purity of 80%.

Embodiment 2

[0029] Wash and dry 500 g of crude drug Scrophulariaceae Scrophulariae, crush, soak in 5 liters of deionized water at room temperature for 36 hours, and filter. The residue was soaked in 2.5 liters of deionized water for 12 hours, filtered, and the combined filtrates were concentrated to 2 liters under reduced pressure at 50°C. Then precipitate with 95% acetone, the final concentration of acetone is 82%, and precipitate for 48 hours. Centrifuge, dissolve the precipitate with 2 liters of water, centrifuge, and dialyze with deionized water for 72 hours. Concentrate under reduced pressure at 50°C and freeze-dry to obtain 35 g of total sugar from Scrophulariaceae. Add 2 liters of water to dissolve, add active carbon to treat, remove repeatedly three times, then concentrate under reduced pressure, centrifuge, and freeze-dry to obtain 17 g of total sugar of Scrophulariaceae Scrophulariae with a purity of 80%.

Embodiment 3

[0031]Scrophulariaceae total sugar was purified by DEAE-Cellulose column chromatography with a purity of 85-90%, and then SephadexG-50 column chromatography and Sephadex G-10 desalted to obtain Scrophulariaceae oligosaccharide SnS-2 with a purity of 98% . Specific separation method: weigh 100 mg of total sugar from Scrophulariaceae, dissolve in 2-3 mL of distilled water, and centrifuge (4500 r / min, 10 min). Put the supernatant on a glass column (1.5cm×60cm), elute with 0.1M NaCl at a flow rate of 0.5mL / min, collect 3-5mL in each tube, and about 11-17 tubes are the absorption peaks. After collection, it was desalted with Sephadex G-10 and freeze-dried to obtain 60 mg of pure oligosaccharide SnS-2.

Abstract

A process for extracting oligomeric saccharide SnS-2 from scrophularia root includes soaking in water, precipitating in organic solvent, dissolving precipitate in water, dialyzing supernatant, membrane separation, concentrating and drying. Its advantages are sure medical action for suppressing cancer and improving immunity, high yield and no toxic by-effect.

Description

technical field [0001] The invention relates to a homologous mixture of oligosaccharides extracted from Chinese medicinal materials, in particular to crocus oligosaccharides extracted from crude drug Scrophulariaceae Scrophulariae, its production method and application. technical background [0002] Scrophulariaceae is a traditional Chinese medicinal material, which has the effects of "cooling blood and nourishing yin, reducing fire, promoting body fluid, and detoxifying". Literature reports, [Zhang Wenjie et al., Yunnan Botanical Research, 16(4), 407(1994)], [Li Yiming, Jiang Shanhao, Gao Wenyun, Zhu Dayuan, Acta Pharmaceutica Sinica, 34(6), 448(1999)], [Zou Chenting, Yang Xiuwei, Chinese Herbal Medicine, 3l(4), 241(2000)], its chemical components mainly include alkaloids, flavonoid glycosides, sterols, amino acids, fatty acids, volatile oils, carotene, iridoid glycosides and trace elements, etc. Studies on sugars in Scrophulariaceae have not been reported yet.
