Embodiment 1

[0025] 1. Total RNA extraction and cDNA synthesis of adult white moths of different sexes

[0026] White moths fed by adults of different sexes were collected. Each treatment had three biological replicates, and they were frozen in liquid nitrogen. After the treatment, total RNA was extracted by Trizol method, and 5 mL of RNase-free DNase I (TakaraBio Inc. , Kusatsu, Shiga Prefecture, Japan) for 15 minutes to remove DNA. The concentration and quality of RNA were detected using a ThermoScientific NanoDrop 2000 UV-Vis spectrophotometer (Thermo Fisher Scientific Inc., Waltham, MA, USA). The integrity of total RNA was detected by 1.0% agarose gel electrophoresis. Using PrimeScript TM II first-strand cDNA synthesis kit (TaKaRa, Dalian, China), purify RNA and synthesize the first-strand cDNA according to the instructions. Perform reverse transcription in a 20.0 μL reaction system containing 4.0 μL 5XPrimeScript IV cDNA Synthesis Mix, 1.0 μL random primer (50 μM), 4.0 μL total RNA...

Abstract

The invention discloses fluorescent quantitative reference genes of hyphantria cunea adults with different sexes as well as primers and application of the fluorescent quantitative reference genes, and belongs to the field of molecular biology of forestry insects. The fluorescent quantitative reference genes are RPL13 and RPS15 genes, and the nucleotide sequences of the fluorescent quantitative reference genes are shown as SEQ ID NO. 1 and SEQ ID NO. 2. According to a transcriptome sequencing database of hyphantria cunea, eight candidate reference genes commonly used as reference genes are selected, the stability of the candidate reference genes is evaluated through five algorithms, the fluorescent quantitative reference genes of hyphantria cunea adults with different sexes are obtained, the real-time fluorescent quantitative PCR primers of the reference genes are designed, and the primers are high in specificity, and high in amplification efficiency. The current situation that the hyphantria cunea adults with different sexes have no suitable reference genes is filled up, and a reliable analysis basis is provided for expression quantification of related functional genes of the hyphantria cunea under different sex experiment conditions.

Description

technical field [0001] The invention belongs to the field of forestry insect molecular biology. It specifically relates to an internal reference gene for fluorescence quantification of adult white moths of different sexes, primers and applications thereof. Background technique [0002] The American white moth (Hyphantria cunea (Drury)) belongs to Lepidortera of Lepidoptera and Arctiidae of the family Arctiidae. It is a worldwide quarantine pest and is listed as one of the first invasive alien species in my country. It has a wide range of hosts, and has the characteristics of miscellaneous food habits, fast transmission, and rampant harm. It mainly harms fruit trees, street trees and ornamental trees, especially broad-leaved trees, including sugar maple, mulberry, elm, ailanthus, willow, hawthorn, apricot, and sycamore. It is harmful to eat leaves, which seriously affects the normal growth of plants, fruit and vegetable yield and quality. At the same time, it will cause se...
