[0130] 1. Materials and methods

[0131] 1. Tissue samples and database

[0132] This example includes glioma samples and normal brain tissues from the Neurosurgery Department of Qilu Hospital of Shandong University. The study protocol was approved by the institutional review board of the hospital, and written informed consent was obtained from each subject. Glioma specimens were validated and classified according to the WHO tumor classification by two experienced clinical pathologists. 5 cases of normal brain tissue, 11 cases of WHO grade II-III, and 18 cases of tissue samples of IV grade (GBM) were obtained from patients with conventional surgical treatment. The clinical information and expression data of glioma samples in public databases were obtained from databases such as TCGA and CGGA.

Abstract

The invention provides an application of LIGHT to diagnosis and treatment of glioma, and belongs to the technical field of crude drugs and molecular biology. Through researches, the inventor finds that the expression quantity of the LIGHT is related to multiple molecular pathology indexes of the glioma, and the malignant progress of the glioma can be effectively inhibited by specifically knockingout the LIGHT. Therefore, the LIGHT can be used as a molecular biomarker for diagnosing and treating the glioma, and can be used as a prognosis index and a potential treatment target of the glioma, sothat the LIGHT has good practical application value.

Description

technical field [0001] The invention belongs to the technical field of crude drug medicine and molecular biology, and specifically relates to the application of LIGHT in the diagnosis and treatment of glioma. Background technique [0002] The information disclosed in this background section is only intended to increase the understanding of the general background of the present invention, and is not necessarily taken as an acknowledgment or any form of suggestion that the information constitutes the prior art already known to those skilled in the art. [0003] Glioma is the most common primary tumor of the central nervous system, accounting for 32% of all primary brain tumors, of which more than half are the most malignant WHO grade IV glioblastoma (Glioblastoma multiform, GBM). Studies have shown that the overall survival of about 88% of GBM patients after diagnosis is less than 1 year, and the 5-year survival rate is <5%. The main reasons for the poor prognosis of GBM ...
