Abstract

The present invention provides a tissue culture and rapid propagation method of iris japonica 'Butterfly Dreams' and belongs to the field of botany. The tissue culture and rapid propagation method ofiris japonica 'Butterfly Dreams' comprises the following steps: (1) disinfection of explants; (2) induction of callus; (4) differentiation of the callus into cluster buds; (5) subsequent proliferationof the cluster buds; (6) seedling strengthening and rooting of the cluster buds; and (7) tissue culture seedling exercising and transplanting. Young and tender flower stems of the iris japonica are used as the explants, the callus is differentiated to form seedlings by indirect induction which is faster than a direct induction for propagation, a large number of new plants are obtained in a shortperiod of time to maintain excellent shapes of a parent plant and can be used as a transgenic receptor system, and the method lays a foundation for further popularization and application of the variety and transgenosis research.
