Embodiment Construction

[0039] The present invention will be described in further detail below in conjunction with the accompanying drawings and embodiments.

[0040] 1. Immunogen preparation

[0041] (1) Synthesis of sialic acid immunogen (SA-BSA) by mixed anhydride method

[0042] Weigh 20 mg of sialic acid and dissolve it in 10 mL of PBS buffer (0.01 mol / L, pH 7.2-7.4), then add 30 μL of triethylamine and 30 μL of isobutyl chloroformate, and react at room temperature for 30 minutes to obtain solution A. Weigh 20 mg of BSA and dissolve it in 1 mL of PBS buffer (0.01 mol / L, pH 7.2-7.4) to obtain liquid B. Slowly add liquid A to liquid B, and shake and react for 24 hours at room temperature in the dark. After the reaction was completed, the resulting reaction solution was transferred to a dialysis bag (1 kD) cleaned with distilled water, and dialyzed in distilled water at 4°C for 72 hours, with the distilled water replaced 3 to 6 times in the middle. The dialyzed product was subpackaged and freeze...

Abstract

The invention discloses a method for rapidly detecting an influenza susceptible population by using a sialic acid artificial antigen and a test strip. In the sialic acid artificial antigen, carrier protein is bovine serum albumin or ovalbumin; animal is immunized with the artificial antigen to obtain a corresponding antibody, which specifically binds to sialic acid in a human saliva sample. A colloidal gold immunoassay test strip which is prepared from the above antibody and a coating antigen is established in the invention, and is used for detecting sialic acid in human saliva samples to quickly determine the influenza susceptible population. The test strip of the invention has high sensitivity and strong specificity and is convenient use, and the detection result is easy to observe and distinguish. The test strip is suitable for rapid clinical measurement and large-scale application at the base level.

Description

technical field [0001] The invention relates to the technical field of biochemical industry, in particular to a method for rapidly detecting influenza-susceptible people by using sialic acid artificial antigen and a test strip. Background technique [0002] Influenza, referred to as influenza, is an acute respiratory infectious disease caused by influenza virus, which can cause multiple complications such as myocarditis, pneumonia, and bronchitis. Because influenza viruses are highly contagious, epidemics, and even worldwide pandemics, can easily occur. Influenza viruses belong to the Orthomyxoviridae family, and the virus particles are spherical and consist of an outer membrane and a nucleocapsid surrounded by it. There is an important structural protein on the surface of the outer membrane, which exists in the form of H polymers, called hemagglutinin (HA). The way influenza virus invades the human body is mainly through the HA on its surface and the terminal glycoprotein ...
