Embodiment 1

[0053] A method for promoting the differentiation of human pluripotent stem cells into neurons, comprising the following steps:

[0054] (1) Human pluripotent stem cells were cultured using a feeder-free method until the cells proliferated to a passable density.

[0055] (2) Digest the human pluripotent stem cells obtained in step (1) with enzymes, and then replace the culture medium with NIM on the same day, which is the 0th day of cell differentiation.

[0056] (3) Human pluripotent stem cells could be observed to form Embryonic bodies (EBs) on the first day.

[0057] (4) On the 7th day, the EBs were adhered to the wall.

[0058] (5) From the 10th day, cells can be observed to form a rosette-like (Rossete) neural tube-like structure.

[0059] (6) The neural tube-like structure formed was blown down on the 16th day, and the formation of neural precursor cells could be observed on the next day.

[0060] (7) On the 18th day, the culture medium was replaced with NeuronM conta...

Abstract

The invention discloses a method for promoting human pluripotent stem cells into mature neurons through directional differentiation. Through a neural induction and a neural differentiation medium, thehuman pluripotent stem cells differentiate into the neurons. By means of the method, time needed for mature neuron obtained through differentiation can be obviously shortened; compared with the priorart, the method has the advantages that the human pluripotent stem cells can be induced to become the mature neurons in vitro in a short time, so that time consumption of neural differentiation is greatly reduced, and a cell model is provided for cell transplantation treatment, drug screening and pathological mechanism study of neurodegenerative diseases.

Description

technical field [0001] The invention belongs to the field of biomedicine, and in particular relates to a method for promoting directional differentiation of human pluripotent stem cells into mature neurons. Background technique [0002] Diseases of the central nervous system are the main diseases that threaten human beings at present, seriously affecting people's quality of life, such as Parkinson's disease, epilepsy, and lateral sclerosis. Because of the characteristics of high incidence, high mortality and high disability rate, these central system diseases bring heavy burden to society and family. [0003] The main pathological change of Parkinson's disease is the loss of dopaminergic neurons in the brain due to extensive degeneration and death, especially in the substantia nigra compacta and striatum; epilepsy is the sudden abnormal discharge of neurons, leading to transient impairment of brain function A chronic relapsing sexually transmitted disease; amyotrophic later...
