Embodiment 1

[0026] The β-cyclodextrin dialdehyde / voriconazole inclusion compound prepared by freeze-drying method was used as a cross-linking agent to cross-link collagen to prepare a collagen membrane material containing antifungal drugs. The preparation steps of this collagen membrane material are as follows:

[0027] (1) Add β-cyclodextrin dialdehyde into deionized water and stir at 60°C to form a saturated solution, then add voriconazole (molar ratio 1:1) after stirring, fully react at constant temperature for 2 hours, freeze-dry, dissolve in acetone, and pump Filter and dry to obtain β-cyclodextrin dialdehyde / voriconazole inclusion compound (β-CD-DA / Vor);

[0028] (2) Freeze-dry the type I collagen extracted from bovine Achilles tendon, and dissolve it with acetic acid or hydrochloric acid to prepare a collagen solution with a concentration of 4-8 mg / mL;

[0029] (3) Add the clathrate obtained in step (1) into the collagen solution as a cross-linking agent. The amount of the cross-l...

Embodiment 2

[0033] The β-cyclodextrin dialdehyde / voriconazole inclusion compound prepared by saturated aqueous solution was used as a cross-linking agent to cross-link collagen, and a collagen membrane material containing antifungal drugs was prepared. The preparation steps of this collagen membrane material are as follows:

[0034] (1) Add β-cyclodextrin dialdehyde to deionized water to make a saturated solution at 30°C, stir and add voriconazole (molar ratio 1:1), keep it still at constant temperature for 1 hour, and filter off the supernatant , suction filtration and drying to obtain β-CD-DA / Vor clathrate;

[0035] (2) Freeze-dry the type I collagen extracted from bovine Achilles tendon, and dissolve it with acetic acid or hydrochloric acid to prepare a collagen solution with a concentration of 8 mg / mL;

[0036] (3) Add the clathrate obtained in step (1) into the collagen solution as a cross-linking agent, the amount of the cross-linking agent accounts for 5% of the collagen mass, sti...

Embodiment 3

[0040] The β-cyclodextrin dialdehyde / voriconazole inclusion compound prepared by ultrasonic method was used as a cross-linking agent to cross-link collagen to prepare a collagen membrane material containing antifungal drugs. The preparation steps of this collagen membrane material are as follows:

[0041] (1) Add β-cyclodextrin dialdehyde to deionized water at room temperature and ultrasonically make a saturated solution, then add voriconazole (molar ratio 1:0.5) ultrasonically, react at a constant temperature for 60 minutes, let it stand still, filter the supernatant, suction filter, Dry to obtain β-CD-DA / Vor clathrate;

[0042] (2) Freeze-dry the type I collagen extracted from bovine Achilles tendon, and dissolve it with acetic acid or hydrochloric acid to prepare a collagen solution with a concentration of 4 mg / mL;

[0043] (3) Add the clathrate obtained in step (1) into the collagen solution as a cross-linking agent, the amount of the cross-linking agent accounts for 30% ...

Abstract

The invention discloses a preparation method for an antifungal cornea repairing material. The preparation method comprises the following steps: (1) preparing a saturated solution of dialdehyde beta-cyclodextrin, adding a medicine and carrying out a full reaction so as to obtain a dialdehyde beta-cyclodextrin / medicine inclusion complex; (2) extracting I type collagen, subjecting the extracted I type collagen to freeze-drying and dissolving the freeze-dried I type collagen in an acetic acid or hydrochloric acid solution to prepare a collagen solution; (3) adding the inclusion complex obtained inthe step (1) as a cross-linking agent into the collagen solution to remove bubbles in the solution; and (4) casting the cross-linked collagen solution to a cornea-repairing-material forming die for film formation. The material prepared by using the method has high transparency, good mechanical properties, and temperature-sensitive release properties, is obviously different in releasing effect atdifferent temperatures, and can be used for different medical fields.

Description

technical field [0001] The invention relates to the field of materials, in particular to a preparation method of an antifungal corneal repair material. Background technique [0002] The cornea is the transparent tissue that sits on the outermost layer of the eyeball and is susceptible to infection and trauma. Corneal diseases (such as leukoplakia, keratoconus, corneal ulcer or trauma, etc.) make the cornea change from transparent to cloudy and affect the patient's vision. Corneal transplantation is one of the effective ways to treat corneal diseases, and allograft transplantation has the best effect. It is currently the first choice for the treatment of corneal ulcers, corneal opacity and other diseases. Due to the limitation of allogeneic corneal donors, corneal repair materials have become a research hotspot. [0003] Worldwide, especially in developing countries, infectious keratitis is the leading cause of blindness next to cataract. Among infectious corneal diseases,...
