Embodiment 1

[0025] A method for visual and rapid detection of methylmercury compounds, comprising the following steps:

[0026] 1) Establishment of comparison standards: 20 μL, 75 μM HT9 DNA aptamer sequence, 10 μL, 0.2 mM silver nitrate and a series of 10 μl of 0 μM, 10 μM, 20 μM, 50 μM, 100 μM, 200 μM methylmercury standard solutions were thoroughly mixed After that, place it at 0°C for 1 hour; then add 150 μL, 10 mM, pH=8.0 Tris-HNO to the above solution 3 Buffer solution, mix well and cool down to 0°C, then add 5 μL, 25mM NaBH 4 Solution, mix well and centrifuge for 10 seconds, then keep it at 25 ℃ for 8 minutes; then add 5 μL 2% HAuCl 4 Solution, after mixing and centrifuging for 10 seconds, keep the temperature at 95°C for 10 minutes, and use a digital camera to record the color change of the solution (as the concentration of MeHg increases, the color of the solution gradually changes from colorless to purple, from light to dark), Corresponding to the concentration of organic merc...

Abstract

The invention belongs to the technical field of detection of organic mercury in a water environment and water products and particularly relates to a method for visually and rapidly detecting an organic mercury compound. The method comprises the following steps: firstly, sufficiently and uniformly mixing an HT9 DNA aptamer sequence with silver nitrate and organic mercury standard solutions with different concentrations respectively; secondly, separately and subsequently adding a Tris-HNO3 buffer solution, a NaBH4 solution and an HAuCl4 solution and then carrying out constant-temperature operation at the temperature of 95 DEG C for 10 minutes, and recording the color change of the solution by using a digital camera, and enabling the color change to correspond to the concentration of the organic mercury solution, thus establishing a standard color card; thirdly, operating to-be-tested liquid according to previous steps, recording the color of the solution by using the digital camera or observing the color of the solution with naked eyes, and comparing the color of the solution with the standard color card, thus realizing the semi-quantitative detection of the organic mercury in a water sample or an aquatic product extracting solution. The method has the advantages of simple operation, high detection speed (the whole detection process is completed within 2 hours), as well as low detection cost (the detection cost of a sample is lower than 2 yuan).

Description

technical field [0001] The invention belongs to the technical field of detecting organic mercury in water environment and aquatic products, and in particular relates to a method for visually and rapidly detecting organic mercury compounds. Background technique [0002] Mercury (Hg) is the only liquid metal at room temperature and is highly toxic. Mercury poisoning can cause irreversible damage to the human body. Inorganic mercury (Hg2+) released into the environment is prone to bioreactions and speciation changes to form organic mercury such as methylmercury (MeHg) and ethylmercury (EtHg). Therefore, there are many different forms of mercury compounds in the environment at the same time, including inorganic mercury (Hg 2+ ), methylmercury (MeHg) and ethylmercury (EtHg). Mercury exists in different forms in the environment, and its bioavailability, mobility and toxicity are quite different. Generally, the toxicity of organic mercury is much higher than that of inorganic ...
