Problems solved by technology

Guar gum has good water solubility and thickening properties, but the original powder often has the following disadvantages: slow dissolution rate, unable to quickly swell and hydrate; difficult to control viscosity; poor thermal stability, etc.
Oxidative degradation is a commonly used method to reduce the viscosity of colloids in chemical degradation, but chemical reagents are easy to cause residues, and the reaction is more violent and difficult to control
Enzymatic reaction conditions are mild, but too high substrate concentration results in high viscosity of the reaction system, slow reaction time, and incomplete enzymatic hydrolysis, while low substrate concentration will cause waste of time and resources in industrial production

Abstract

The invention provides a method for preparing galacto-mannan-oligosaccharides (GMOS). The method comprises the following steps: with guar gum as a substrate, compounding and degrading with hydrogen peroxide and ascorbic acid; removing residual hydrogen peroxide in the system by utilizing catalase, and adding commercial acidic beta-mannanase for performing enzymolysis; performing enzyme deactivation on hydrolysate, centrifuging to remove impurities, concentrating, precipitating to remove carbohydrates of large molecular weight by utilizing 3-5 times ethanol, performing alcohol precipitation on supernatant, performing suspended evaporation and concentrating to the solid content of more than or equal to 20%, performing spray drying, thereby obtaining GMOS dried powder with the molecular weight of less than 10000Da; and performing wet granulation on GMOS dried powder with 80%-95% thanol, thereby obtaining solid particles with the granularity of 40-100 meshes and high solubility.

Description

technical field [0001] The invention belongs to the technical field of biological product preparation, and in particular relates to a preparation method of galactomannan oligosaccharides. Background technique [0002] Guar (Cyamops tetragonooba (L.) Taub.) is a tropical leguminous herb widely planted in India, Pakistan and other places. Guar gum (Guar gum) is obtained by peeling the endosperm part of guar seeds after removing the germ, drying and pulverizing, adding water, carrying out pressurized hydrolysis, precipitating with 20% ethanol, drying and pulverizing after centrifugation. The viscosity is the highest among natural colloids, and it is one of the cheapest and most widely used hydrophilic colloids in the world. Its main chemical composition is galactomannan, the main chain is (1→4)-β-D-mannose unit, and the side chain is a single α-D-galactose with (1→6) bond with the main chain. The ratio of galactose to mannose is 1:1.8, which is about 1:2. Guar gum has good w...
