Problems solved by technology

[0005] Mycoplasma pneumoniae can be cultured in vitro with PPLO medium, so the immunofluorescence method and enzyme-linked immunoassay method for detecting Mycoplasma pneumoniae antibodies have been established by using the cultured Mycoplasma pneumoniae, but it is difficult to mass-produce the antigen detection antibody using the cultured Mycoplasma pneumoniae False positives are generated, and the development of specific genetic recombinant antigens is the development direction of the establishment of Mycoplasma pneumoniae specific antibody detection reagents
The most ideal way to prevent Mp infection is to inject vaccines, but there is no vaccine to prevent Mp at present, and the research and development of vaccines are being actively carried out at home and abroad, especially the research on inactivated vaccines is progressing rapidly, and genetic engineering vaccines are the ultimate development direction

Embodiment Construction

[0200] Detailed description of the embodiment of the present invention:

[0201] Mycoplasma pneumoniae P116 protein, P1 protein and P30 Protein epitope analysis, gene synthesis and expression

[0202] Analyze the amino acid sequence of Mycoplasma pneumoniae P116 protein, P1 protein and P30 protein by computer, and screen out the P116 protein fragment of Mycoplasma pneumoniae containing a strong epitope, that is, the 261st amino acid to the 466th amino acid, and Mycoplasma pneumoniae containing a strong antigenic epitope The P1 protein fragment of P1 protein, that is, the 1125th amino acid to the 1395th amino acid, and the P30 protein fragment of Mycoplasma pneumoniae that contains a strong epitope, that is, the 107th amino acid to the 161st amino acid.

[0203] Select codons preferred by both eukaryotes and prokaryotes, chemically synthesize a new tandem gene sequence of Mycoplasma pneumoniae P116 protein fragment, P1 protein fragment and P30 protein fragment, and u...

Abstract

The invention discloses a fusion protein of a mycoplasma pneumonia protein epitope and preparation and application thereof, and relates to the fields of genetic engineering technologies, vaccines and diagnosis reagents. Amino acid sequences of a mycoplasma pneumoniae P116 protein, a P1 protein and a P30 protein are analyzed through a computer, mycoplasma pneumoniae P116 protein fragments, namely from the 261st amino acid to the 466th amino acid containing the strong epitope, mycoplasma pneumoniae P1 protein fragments, namely from the 1125th amino acid to the 1395th amino acid containing the strong epitope and mycoplasma pneumoniae P30 protein fragments, namely from the 107th amino acid to the 161st amino acid containing the strong epitope are screened out. Series connection gene sequences of novel mycoplasma pneumoniae P116 protein fragments, mycoplasma pneumoniae P1 protein fragments and mycoplasma pneumoniae P30 protein fragments are synthesized chemically, three gene fragments are connected in series, the fusion protein of P116 protein fragments, P1 protein fragments and P30 protein fragments is expressed by means of the genetic engineering technology, the three protein fragments are connected through glycine-glycine-serine, and 538 amino acids are formed in the overall length of the fusion protein.

Description

technical field [0001] The fusion protein of the mycoplasma pneumoniae protein antigen epitope and its preparation and application in the present invention relate to the fields of genetic engineering technology, vaccines and diagnostic reagents. The amino acid sequence of the mycoplasma pneumoniae P116 protein, P1 protein and P30 protein is screened out by computer. The P116 protein fragment of Mycoplasma pneumoniae with a strong antigenic epitope, the P1 protein fragment of Mycoplasma pneumoniae with a strong antigenic epitope, and the P30 protein fragment of Mycoplasma pneumoniae with a strong antigenic epitope. Select codons favored by both eukaryotes and prokaryotes, chemically synthesize a new tandem gene sequence of Mycoplasma pneumoniae P116 protein fragment, P1 protein fragment and P30 protein fragment, and express P116 protein fragment, P1 protein fragment and P30 protein fragment by genetic engineering technology The fusion protein, the three protein fragments are co...
