Embodiment Construction

[0026] The invention detects the single nucleotide polymorphism of the 12265th mutation of the yellow cattle Leptin gene, so as to be used for marker-assisted selection of growth traits of Chinese yellow beef, and quickly establish a yellow cattle population with excellent genetic resources. In the following, the present invention will be further described in detail in combination with the detection of specific samples and the association of traits, which is an explanation of the present invention rather than a limitation.

Abstract

The invention discloses a method for detecting single nucleotide polymorphism of cattle Leptin genes. According to the method, whole-genome DNA containing the cattle Leptin genes to be detected is used as a template, a primer pair P is used as the primer, a PCR method is adopted for amplifying a second exon segment of the cattle Leptin genes, the restriction enzyme Pst I is used for digesting a PCR amplified product, the enzyme-digested segment is subjected to polyacrylamide gel electrophoresis, and the single nucleotide polymorphism of the site 12265 of the cattle Leptin genes is authenticated according to an electrophoresis result. Since the Leptin genes have an important biological function on controlling animal growth, fat differentiation and meat formation, the detection method lays a foundation for establishment of association between SNPs and growth traits of the Leptin genes. It is also found that the site 12265 (T / C) of the Leptin genes has a remarkable influence on early growth traits, including birth weight, body weight and chest circumference, of Nanyang cattle.

Description

technical field [0001] The invention belongs to the field of molecular genetics and relates to the detection of single nucleotide polymorphism (Single Nucleotide Polymorphism, SNP), in particular to a method for detecting the 12265th single nucleotide polymorphism of cattle Leptin gene. Background technique [0002] Single nucleotide polymorphism (SNP) refers to the polymorphism caused by the substitution of a single nucleotide (A / T / C / G) in the genomic DNA sequence. Generally speaking, SNPs include base substitution, Insertions, deletions, and copy number changes of repetitive sequences. In diploid organisms, SNPs can theoretically be composed of 2, 3 or 4 alleles, but in fact, SNPs with 3 or 4 alleles are very rare, so SNPs are usually called biallelic Gene molecular markers. According to the distribution position of SNPs in the genome, it can be divided into gene coding region SNPs (cSNPs), gene peripheral SNPs (pSNPs) and intergenic SNPs (iSNPs). Among them, cSNPs have...
