Problems solved by technology

However, the envelope protein gene of the pseudovirus reported in the literature is mainly derived from the sequence of the standard strain, the detection specificity is limited, and the packaging efficiency of the pseudovirus is low, which cannot meet the follow-up experiments such as neutralizing antibody evaluation (vaccine evaluation)
In addition, there are obvious differences in the packaging efficiency of different subtypes of pseudoviruses. The current research data shows that 5a is the best, and 1b, 2a, and 6a are the worst; while the popular ones in my country happen to be 1b, 2a, and 6a

Embodiment Construction

[0018] The technical solution of the present invention will be further described in detail below in conjunction with the accompanying drawings and specific embodiments.

[0019] refer to figure 1 , the technical solution of the present invention is specifically:

[0020] 1. use LS reagent extracts HCV nucleic acid from the serum of HCV infected patients;

[0021] 2. Cloning and sequence analysis of the envelope protein coding genes of different subtypes of HCV derived from serum and designing general primers;

[0022] 3. RT-PCR accurately amplifies the target gene HCV-CE1E2;

[0023] 4. After gel recovery and purification of the target gene HCV-CE1E2 and TA cloning, dozens of clones were obtained;

[0024] 5. Carry out enzyme digestion to recover the fragments, connect the pVRC expression vector, transform and extract the plasmid, and obtain multiple pVRC-366-CE1E2 expression plasmids;

[0025] 6. pVRC-366-CE1E2 and packaging plasmid pNL4-3.Luc.R-E, using non-liposome tr...

Abstract

The invention discloses an HCV (hepatitis c virus) envelope protein gene and an application. The nucleotide sequence of the HCV envelope protein gene is represented as SEQ ID NO.1. RT-PCR (reverse transcription-polymerase chain reaction) and nested PCR technologies are adopted to obtain HCV 1b subtype envelope protein genes popular in China, an expression vector provided with different HCV 1b subtype envelope protein genes is constructed, a two-plasmid cotransfection method is adopted to establish a pseudovirus database, high-infectivity pseudovirion is screened out, and an effective model is provided for studying screening of anti-HCV medicine and immune effect evaluation of HCV vaccines, meanwhile, early characteristics of HCV infection can be further studied, and a technology platform can be provided for evaluation of response of a serum neutralizing antibody from an HCV patient and an antibody of an experimental vaccine and the like.

Description

technical field [0001] The invention belongs to the technical field of bioengineering, and relates to an HCV envelope protein gene and its application. Background technique [0002] At present, there are about 170 million chronic hepatitis C virus infection patients and patients in the world, accounting for 3% of the global population. Among them, there are about 40 million infected people in China. The biggest feature of hepatitis C is the high rate of slow conversion. About 80% of acute infection patients will develop chronic hepatitis, and about 20% of chronic hepatitis patients will develop liver cirrhosis, and 1% of liver cirrhosis patients- 4% of people will eventually develop liver cancer, so there are currently about 170 million chronic hepatitis C patients in the world who are at risk of developing liver cirrhosis and liver cancer. The worldwide high infection rate, high slow conversion rate and the lack of treatment and vaccine make HCV a research hotspot. [00...
