Problems solved by technology

[0004] It should be further pointed out that for fine systems for detecting blood cells, such as microfluidic platforms and flow cell separation instruments, it is necessary to be able to prevent blood cell aggregation and blood coagulation, that is, to avoid blood sample clogging; however, there is no effective Anticoagulant Preservation Reagent

Embodiment Construction

[0021] The present invention will be described below in conjunction with specific embodiments.

[0022] A reagent for stabilizing cells of a blood sample, comprising materials having the following concentrations, specifically:

[0023] β-cyclodextrin 100μmol / L

[0024] Disodium cromolyn 100μmol / L

[0025] Potassium dichromate 0.05%

[0026] Triflusal 50μmol / L

[0027] Trutroban 100μmol / L

[0028] Sodium citrate 11 mmol / L

[0029] Theophylline 1.5 mmol / L

[0030] Dipyridamole 20μmol / L

[0031] Adenosine 0.37 mmol / L

[0032] Glucose 20 mmol / L

[0033] PEG 1.25 mmol / L.

[0034] For further explanation, the PEG of the present invention is PEG-1000.

[0035] Among them, for the reagents for stabilizing blood sample cells of the present invention, β-cyclodextrin, disodium cromoglycate, and potassium dichromate can directly stabilize blood cell membranes, and triflusal, trutroban, theophylline, dipyrim Damo can effectively protect platelets and inhibit platelet aggregation...

Abstract

The invention discloses a reagent for stabilization of blood sample cells. The reagent comprises 100 micromoles per liter of L beta-cyclodextrin, 100 micromoles per liter of disodium cromoglycate, 0.05% of potassium dichromate, 50 micromoles per liter of triflusal, 100 micromoles per liter of Terutroban, 11 millimoles per liter of sodium citrate, 1.5 millimoles per liter of theophylline, 20 micromoles per liter of dipyridamole, 0.37 millimoles per liter of adenosine, 20 millimoles per liter of glucose and 1.25 millimoles per liter of LPEG. The reagent can stabilize blood cells for at least 36h and prevent blood cell aggregation. Through the reagent, a blood sample subjected to treatment does not influence follow-up sample molecule analysis. The reagent can be widely used for blood sample molecule analysis and detection and is especially suitable for cell or molecular detection such as a microfluidic technology with strict sample cell requirements and an analysis technology platforms such as a flow cytometer with strict sample cell requirements.

Description

technical field [0001] The invention relates to the technical field of blood preservation reagents, in particular to a reagent for stabilizing blood sample cells. Background technique [0002] Stable blood samples are extremely important for the transportation of blood samples and the operation of the clinical trial process, there are some detection techniques and methods, such as enrichment and analysis of rare circulating cells (circulating tumor cells, circulating fetal cells, circulating stem cells and circulating vascular endothelial cells) or The detection of pathogenic bacteria in blood usually requires multi-step and complex enrichment processes. Stabilizing blood cells and enriching target cells that can withstand the enrichment process without cell dissolution are the key to the successful development of such products. [0003] Formalin has been used as a fresh-keeping agent for tissues and cells for nearly a hundred years. Among them, formalin is a preservative th...
