Embodiment

[0084] Establishment of clinical mastitis models of two bacterial types in Chinese Holstein cows No. 9133 (cow 1), No. 9130 (cow 2) and No. 8107 (cow 3)

[0085] In a dairy farm in the suburbs of Yangzhou City, 3 cows were selected that were healthy, had no history of illness, were in normal body condition, had well-developed udders, were in the mid-lactating stage of the first parity, had SCCfigure 1 ), milk bacteriological testing ( figure 2 , image 3 ), breast histopathological section observation ( Figure 4 , Figure 5 , Figure 6 ), all proved that the cow had been infected with clinical mastitis, and the milk bacteriology test showed that the right front (A2) udder of the dairy cow 1 was infected with Staphylococcus aureus, the right rear (A3) udder was infected with Streptococcus, and the left front (A1) ), the left rear (A4) udder had no bacterial infection; the right rear (B3) udder of dairy cow 2 was only infected with Staphylococcus aureus, the left rear (B4) ...

Abstract

The invention provides a method for constructing a clinical mastitis model of two bacterial types in different udder regions of dairy cows. The method comprises the following steps: selecting a Chinese Holstein dairy cow as the experimental animal, wherein the selected dairy cow is healthy, does not have disease history, is normal in body condition, has excellent breast development and is at the middle stage of lactation for the first parity, SCC is less than100000 per mL, and results of milk bacterial culture in all udder regions are negative; respectively carrying out the following treatment on four udder regions of the dairy cow: injecting 1*10<7>CFU / mL of staphylococcus aureus bacterial suspension and 1*10<6> CFU / mL of streptococcus suspension into breasts of the dairy cow via ductus papillaris by using a breast clearing needle, injecting sterile PBS in equal amount for the control group, and not carrying out treatment on a blank group; and reversely pressing the breasts from the base of each nipple from bottom to top with hands in a way that the thumbs and the index fingers are connected into a circle so that the bacterial suspensions can uniformly diffuse to lactiferous ducts and do not accumulate in gland cisterns; and attacking with bacteria for 24 hours to make sure that the dairy cow mastitis model is successfully constructed. According to the method, the influence of individual difference on tests can be completely eliminated.

Description

technical field [0001] The invention belongs to the field of biological sciences, and in particular relates to a method for constructing two bacterial types of clinical mastitis models by using different milk areas of the same dairy cow. Background technique [0002] Dairy cow mastitis, also known as mastitis, is an inflammatory reaction caused by a variety of etiologies. It is one of the most common diseases of dairy cows and is affected by factors such as the genetic background, environmental conditions, and feeding management conditions of dairy cows. [0003] Mastitis has seriously affected the quality of raw milk, the useful life of dairy cows, the benefits of dairy farming, the nutritional value of milk products and the health of consumers. Cow mastitis is one of the main causes of low productivity and high costs in the cattle industry. The incidence of mastitis in dairy cows in most countries is about 40% (including recessive mastitis), and milk production will decre...
