Problems solved by technology

[0004] Although laccase has such an important use, its application in production practice is still very limited. The main reason is that most production strains have low efficiency in synthesizing laccase, resulting in insufficient laccase production.

Embodiment 1

[0075] Implementation 1: Optimization Experiment

[0076] Using the optimized medium formula and fermentation conditions to ferment and produce enzymes, the highest laccase activity can reach 58888.59U / L, which is about 10.8 times that before optimization.

Embodiment 2

[0077] Implementation 2: 5L fermenter test

[0078] An automatic control 5L fermenter was used, with a liquid volume of 3.2L, an inoculum volume of 8%, and a fermentation temperature of 25°C. The fermentation liquid was taken at 8000r / min and centrifuged at 4°C for 10min every day, and the supernatant was taken to determine the laccase activity (Table 13).

[0079] Table 13 Changes of laccase activity in the fermentation process of Dendrobium unicolor

[0080]

[0081] With the prolongation of fermentation time, the activity of laccase also gradually increased. When the fermentation reached the 6th day, the enzyme activity reached the maximum, which was 79733U / L, and the highest enzyme activity was about 1.35 times that under the laboratory shake flask condition; With the further increase of fermentation time, the enzyme activity began to decrease.

Abstract

The invention provides a method for producing laccase by virtue of fermentation, comprising the following steps: preparing a fungus secretion laccase culture medium, generating enzyme and fermenting, wherein per 100mL of the fungus secretion laccase culture medium comprises 1-4g of glucose, 1-2.5g of ammonium tartrate, 0.05-0.25mmol / L of ABTS, 10.6mg / L of VB, macroelements and trace elements, andthe balance of water; and adjusting the initial pH value to be 5.0-6.0 by utilizing NaOH or HCl. Along with the unceasing development of research, the laccase permeate can be applied to the fields ofindustry, agriculture and medical treatment and the like, however, the reports in the aspects of large-scale industrial production of the laccase and exploration of the proper fermentation condition and the like are less, which is still the hot spot in the future research. By the method provided by the invention, the process condition for producing the laccase by virtue of fermentation is established, and the output and quality of the laccase can be improved; and the method lays a foundation for realizing the industrial production of the laccase.

Description

technical field [0001] The invention belongs to the field of biomanufacturing, and more specifically relates to a method for fermenting and producing laccase by using Dendrobium monochrome. Background technique [0002] Lignin, cellulose and hemicellulose are the three most abundant renewable resources in nature, accounting for 20%-35%, 45% and 20% of the dry weight of plants respectively. Lignin is a natural polymer compound widely present in the cell walls of higher plants. It is a biopolymer whose reserves are second only to cellulose, and is regenerated at a rate of more than 5 billion tons per year. The complete degradation of lignin in nature is mainly accomplished through the decomposition of white rot fungi. Its mechanism is realized by extracellular oxidases secreted by white rot fungi—lignin degrading enzymes, among which laccase ( Laccase ), manganese peroxidase ( MNP ) and lignin peroxidase ( LiP ) is the main component of the lignin-degrading enzyme system...
