Embodiment 1

[0026] 1. Isolation of Serratia marcescens GEI

[0027] Healthy Chinese honey bees were purchased from Conghua District, Guangzhou City, Guangdong Province.

[0028] (1) Retrieve healthy adult worker bees from the middle bee colony.

[0029] (2) Soak the worker bees in 75% (v / v) alcohol for 10 seconds in an ultra-clean workbench, and then sterilize them in 0.1% mercury chloride solution (w / v) for 2 minutes. Worker bees were washed with sterile water and then dissected in a sterile Petri dish.

[0030] (3) Hold the poisonous needle at the tail of the bee with tweezers, and carefully drag out the midgut.

[0031] (4) Grind the midgut in a sterilized mortar, and dilute the grinding solution to 10% with sterilized water. -1 -10 -8 , take 10 -6 、10 -7 、10 -8 3 dilutions.

[0032](5) Take 100 μL of each dilution and spread them on LB medium and PDA medium respectively, and each dilution is repeated 3 times.

[0033] (6) Culture the culture medium at 28°C for about 2-3 days,...

Abstract

The invention discloses a chitinase for deactivating a varroa destructor, a serratia marcescens and application thereof. In the invention, the serratia marcescens for deactivating the varroa destructor is separated from intestines of Chinese worker bees and is stored in a Chinese typical culture storage center with a storage number of CCTCC M 209097; a gene base sequence is cloned from the serratia marcescens, and a procaryon expression carrier pET-32a(+) is applied to express the gene in a colon bacillus. Purification, renaturation and bioassay are carried out on a protein coded by the gene,a result shows that the protein coded by the gene has stronger toxicity to the varroa destructor, and sequencing analysis shows that the protein is a new sequence of chitinase gene ChiA types. The invention can be used for preventing the mite damage of bees.

Description

Technical field: [0001] The invention belongs to the field of biotechnology, and in particular relates to chitinase ChiA and Serratia marcescens which have the activity of killing Varroa mite, and their application in killing Varroa mite. Background technique: [0002] Bee parasitic mites have always been one of the most important hazards to the world's beekeeping industry. They are harmful on a global scale. At present, except for Australia and some parts of Africa, bee mites have not been found, and bee colonies in other continents are harmed by bee mites. The ectoparasitic mite Varroa destructor Anderson&Trueman (Varroa destructor Anderson&Trueman) sucks the body fluids of bee adults or larvae, causing the decline of bee physique and the weakening of bee colonies; in addition, bee mite also spreads bee bacteria and viruses disease. According to reports, the honeybee colony collapse disorder (CCD) that recently appeared in the United States and other countries is also rel...
