Example 1: Enteric coated glucosinolate and myrosinase granules

A) Formation of glucosinolate and β-thioglucosidase granules

[0059]The preparation of glucosinolate and β-thioglucosidase granules can generally be performed using four sequential steps. The first of these steps is formulation and formation of a compressible mixture. After this step, the mixture is fed to a pelletizer / extruder to produce a compressed extrudate. The resulting extrudate is then fed to a spheronizer to form glucosinolate or β-thioglucosidase granules. The granules are recovered and then dried in a fluidized bed.

[0070] Using ProCoater from Torpac (Fairfield, N, J.) with EUDRAGIT from Degussa Coating or Cellulose Acetate Phthalate ("CAP") from Eastman Chemical Co. (Kingston, T.N.) Gelatin capsules from Wonder Laboratories (White House, TN) were coated. EUDRAGIT And CAP coating can resist gastric acid. Typically, a coating is applied to provide about 8 to about 12% coating. Glucosinolates and β-thioglucosidase were stable during coating as determined by HPLC. The coated capsules were tested according to the Torpac method (Fairfield, NJ) and remained stable in 0.1N HCl at 37°C for more than 2 hours, but completely soluble in phosphate at pH 6.8 within 30 minutes at 37°C in the buffer.

Abstract

The present invention relates to a particulate composition comprising enteric-coated glucosinolate and beta-thioglucosidase particles. The present invention further provides a method of converting glucosinolate to isothiocyanate in the small intestine comprising orally administering to a subject an enteric-coated chemoprotectant precursor composition comprising enteric-coated glucosinolate and beta-thioglucosinodase particles. In another aspect, uncoated glucosinolate and beta-thioglucosinodase particles may be provided in an enteric-coated capsule. Preferably, the glucosinolate is glucoraphanin and the beta-thioglucosidase is myrosinase. The enteric coating targets the compound for release in the small intestine where beta-thioglucosinodase enzyme converts glucosinolate to chemoprotectant isothiocyanate.
