Embodiment 1

[0037] 1) Preparation of gold nanorods with a length of 70 nm, a diameter of 9 nm, an aspect ratio of 8.3, and a maximum absorption wavelength of 1129 nm.

[0038] Prepare 10mL of 0.1M sodium hexadecylbenzenesulfonate solution as stabilizer, add 500μL of freshly prepared 10mM gold chlorate solution as raw material, stir rapidly, add 20μL of 0.1M silver nitrate solution and stir, then add 20 μL of 1 M hydrochloric acid was used to adjust pH, and finally 500 μL of 0.1 M hydroquinone aqueous solution was added as a reducing agent to prepare gold nanoseeds. The reaction was carried out in a water bath at 30 °C for 15 minutes, and 10 μL of 10 mM sodium borohydride solution was quickly added as an aging agent to promote the longitudinal growth of gold nanoseeds to form gold nanorods. The obtained gold nanorods were 70 nm long, 9 nm wide, and had an aspect ratio of 8.3. The maximum absorption wavelength is in the near-infrared second region at 1129 nm.

Abstract

The invention discloses a cell membrane targeting nano-probe, preparation thereof and application of the cell membrane targeting nano-probe in light response regulation and control of neuron calcium ion flow. According to the invention, the gold nanorod with a small size and a high length-diameter ratio is adopted as a main body structure, the HS-PEG-NHS and the Anti-TRPV-1 antibody are adopted as targeted modification materials, and the assembled probe can be a non-invasive and non-destructive remote safe probe. A gold nanorod which is small in size and high in length-diameter ratio is used as a main body structure, modification of an Anti-TRPV-1 antibody is introduced, the cell membrane targeting characteristic of the nanoprobe is improved, and the nanoprobe can be combined to the TRPV-1 antibody on the surface of a neuron membrane. The characteristic that the TRPV-1 is sensitive to thermal stimulation and the characteristic that the gold nanorod stimulates photothermal conversion of near-infrared light are not combined, a synergistic effect is achieved, under thermal conversion of the gold nanorod, opening and closing of a TRPV-1 channel can be rapidly stimulated, and then neuron calcium ion movement can be reversibly regulated and controlled.

Description

technical field [0001] The invention belongs to the technical field of medical materials, and in particular relates to a cell membrane targeting nano-probe and its preparation and application in regulating the flow of calcium ions in neurons in response to light. Background technique [0002] With the rapid development of nanotechnology, the emergence of nanoprobes provides a more precise intervention strategy for regulating cellular activities. Among them, due to the characteristics of non-invasive, non-destructive and non-invasive, the combination of nanoprobes with optical stimulation to regulate cell life activities has attracted the attention of researchers in the field of life medicine. By adjusting parameters such as the wavelength of light, stimulation time, stimulation duration, and stimulation location, researchers can achieve space-time specific intervention in the life activities of cells, thereby further regulating biochemical changes in the body and assisting d...
