Embodiment 1

[0051] Embodiment 1: the extraction method of Cistanche total glycosides, Cistanche total polysaccharides and Cistanche total oligosaccharides

[0052] Take 5 kg of Cistanche deserticola, add 8 times the amount of water and heat to reflux, each time for 2 hours, extract three times in total, and combine the filtrates. Concentrate the filtrate to a certain volume under reduced pressure, add 95% ethanol until the alcohol content reaches 60%, precipitate at low temperature for 12 hours, filter, and the precipitate is the total polysaccharide of Cistanche cistanche; let it stand overnight; filter, concentrate the filtrate under reduced pressure, and purify it through macroporous resin , followed by elution with water and different concentrations of ethanol. Collect the water eluate, concentrate under reduced pressure to a thick paste to obtain the total oligosaccharides of Cistanche deserticola; collect the 40% ethanol eluate, concentrate under reduced pressure, and dry to obtain ...

Embodiment 2

[0054] Example 2: Pharmacodynamic study of total glycosides of Cistanche deserticola and echinacoside in protecting alcoholic liver injury with intestinal injury

[0055] Thirty male healthy Kunming mice were randomly divided into normal group (NOR), model group (MOD), Cistanche total glycosides group (TGs, 400mg / kg), Cistanche polysaccharide group (PSs, 400mg / kg), and Cistanche oligosaccharide group Group (OSs, 400mg / kg) Six groups, 10 rats in each group. The model of alcoholic liver injury was established by intragastric administration of Red Star Erguotou. Seven days after the establishment of the model, normal saline and drugs were administered by intragastric administration for 14 consecutive days.

[0056] HE staining ( figure 1 ) showed that the liver tissue cells in the NOR group were normal in shape, neatly arranged, and complete in structure; in the MOD group, the structure was disordered, the cells were arranged irregularly, the intercellular space was loose, and t...

Abstract

The invention provides application of cistanche total glycosides and echinacoside in preparation of medicines for preventing and treating alcoholic liver injury with intestinal injury and regulating intestinal flora. Pharmacological experiment results show that in an alcoholic liver injury mouse model, the cistanche total glycosides and echinacoside can relieve mouse liver tissue morphological injury, reduce mouse liver lipid deposition, reduce the content of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) in mouse liver, relieve oxidative injury of mouse liver, and reduce damage of harmful substances such as endotoxin and the like to liver; and meanwhile, the mouse small intestine tissue form damage can be relieved, the intestinal wall villus integrity is improved, and the intestinal flora disorder is regulated. Therefore, the invention has the prospect of being developed into a medicine capable of simultaneously protecting the alcoholic liver injury and the alcoholic intestinal injury, and has important clinical significance and commercial value.

Description

technical field [0001] The invention belongs to the field of medicine, and relates to a new application of Cistanche deserticola extract, in particular to the preparation of total glycosides of Cistanche deserticola and echinacoside in the preparation of drugs for preventing and treating alcoholic liver injury accompanied by intestinal injury and regulating intestinal flora Applications. Background technique [0002] Alcohol consumption is a factor in many disease and injury conditions, and most alcoholics experience tissue damage or organ dysfunction. 20%-30% of heavy alcoholics suffer from alcoholic liver diseases such as alcoholic steatohepatitis and cirrhosis. Chronic alcohol consumption can have damaging effects on growth factors, cytokines and immune function, and damage the body's organs through the inflammatory response. It has been reported that the gastrointestinal tract may be an important source of inflammation in alcohol-mediated organ damage. [0003] Alcoho...
