Problems solved by technology

[0005] However, the existing solid-phase gene chips, such as the sheep Illumina 50K chip, have the following problems in the detection process and application: first, there are many typing sites; The SNP sites around these sites cannot be typed, and the SNP sites around these sites cannot be typed. Once the solid-phase chip is designed, the sites that can be detected are fixed, and cannot be added or deleted, and the flexibility is poor; finally , the cost of solid-phase chip typing is high

Embodiment Construction

[0017] The present invention will be described in further detail below in conjunction with the accompanying drawings and embodiments, which are only used to explain the present invention, rather than limit the protection scope of the present invention.

[0018] (1) Design and preparation of sheep 1K liquid phase chip

[0019] The present invention utilizes the resequencing data of a total of 551 sheep (Table 1) and 36 sheep wild relatives (Wild sheep; Table 2) of 64 breeds in the world (including 434 Chinese sheep), and first conducts SNP analysis. After filtering the sites, a total of 99,406,384 SNP sites were obtained for subsequent screening.

Abstract

The invention discloses a sheep liquid phase chip and application thereof, 935 SNP loci for chip design are found and screened, and sheep genotyping can be realized by utilizing the designed liquid phase chip through a targeted capture sequencing technology. Experimental results show that the chip designed by the invention can be used for sheep genetic diversity analysis, variety identification, genetic relationship identification, whole genome association analysis and genome selective breeding.

Description

technical field [0001] The invention relates to the field of whole-genome gene chips, in particular to a sheep liquid-phase chip and its application. Background technique [0002] Molecular Marker Technology is an important tool in molecular breeding. Traditional molecular markers, such as restriction fragment length polymorphism (Restriction Fragment Length Polymorphism, RFLP) and simple sequence repeat (Simple Sequence Repeat, SSR) play an important role in the field of genetic breeding. However, there are also certain limitations, such as the small number of genome distributions, the cumbersome operation process, and low throughput, which make it impossible to meet the needs of large-scale commercial breeding applications. SNP (Single Nucleotide Polymorphism, SNP) refers to the variation of a single nucleotide in the genome, including the conversion, transversion, insertion or deletion of a single base pair. As a more widely distributed genetic marker in the genome, SNP...
