Problems solved by technology

Duscher D et al. reported that systemic injection of transplanted cells has the disadvantages of cell deposition at the wound site, high cell attrition rate, poor positioning, poor tissue targeting, and limited cell survival (see Duscher D, et al. (2016). Gerontology62(2): 216 -225.); Garg RK et al. reported that local injection of cells is widely used in wound therapy, but shear damage during local injection will also reduce the survival rate of injected cells (see Garg RK, et al. (2014).Stem Cells Transl Med 3(9):1079-1089.)

Embodiment Construction

[0056] The technical solutions and beneficial effects of the present invention will be further described below in conjunction with specific embodiments. It should be noted that, in the case of no conflict, the embodiments in the application and the features in the embodiments can be combined with each other.

[0057] Preparation Examples:

[0058] 1. Isolation of human umbilical cord mesenchymal stem cells (hUC-MSCs)

[0059] 1) Rinse the blood in the umbilical cord with normal saline, then cut into small pieces of about 2 cm, completely soak in the "PBS + double antibody" preservation solution, and transport at 4 °C;

[0060] 2) Rinse the disinfectant and blood on the surface of the umbilical cord with PBS (containing 1% double antibody) solution;

[0061] 3) Separate the umbilical artery, the adventitia of the umbilical vein and the surface of the umbilical cord in the PBS solution on the ice table;

[0062] 4) Cut the umbilical cord into 0.5cm pieces, spread them evenly i...

Abstract

The invention provides a human umbilical cord mesenchymal stem cell preparation and application thereof. According to the cell preparation, fibrous protein gel is used as a scaffold during cell transplantation, a three-dimensional environment which is more beneficial to survival is provided for hUC-MSCs cells, and the long-time repair promoting effect can be achieved. The invention further relates to the human mesenchymal stem cells with green fluorescence labels and an application of the human mesenchymal stem cells. Particularly, hUC-MSCs are marked by adopting EGFP, so that in-vivo tracing of transplanted cells and exploration of an action mechanism are realized. In addition, EGFP is adopted for hUC-MSCs, and it is verified that the hUC-MSCs cell preparation provided by the invention can remarkably accelerate healing of diabetic wounds and promote reepithelization.

Description

technical field [0001] The application belongs to the technical field of stem cell application, and in particular relates to a human umbilical cord mesenchymal stem cell preparation and its application; it also relates to a human mesenchymal stem cell with a green fluorescent label and its application. Background technique [0002] Diabetes is a chronic metabolic disease. Its incidence is increasing year by year. It currently affects 9.5% of the world's population. It is estimated that by 2030, the number of people affected may exceed 50% of the global population. Diabetic foot ulcer (Diabetic foot ulcer, DFU) is one of the main complications of diabetes. This injury is associated with diabetic neuropathy and peripheral arterial disease. [0003] It is estimated that every 30s in the world there is an amputation due to diabetic foot (see Boulton AJ, Vileikyte L, Ragnarson-Tennvall G, & Apelqvist J (2005). Lancet (London, England) 366 (9498): 1719-1724.) . However, ischemi...
