Problems solved by technology

[0003] The traditional monitoring process of benthos diversity mainly includes sediment sample collection, manual sample sorting, and morphological identification. high professional requirements
[0006] The above method also has some defects. For example, in the actual water environment, due to the limitation of sampling equipment and shore conditions, it

Embodiment Construction

[0036] The application will be further described below in conjunction with the accompanying drawings. The following examples are only used to illustrate the technical solutions of the present invention more clearly, but not to limit the protection scope of the present application.

[0037] The present invention is a method for monitoring freshwater benthos community diversity based on environmental DNA technology, the method comprising: step 1. collecting surface water samples in the monitoring area; step 2. water sample filtration and eDNA extraction; step 3. PCR amplification Increase; Step 4. High-throughput sequencing to obtain the representative sequence of OTU; Step 5. Establishment of the benthos comparison database; Step 6. Compare the established benthos database, and perform species annotation on the representative sequence of the OTUs of the sample to be tested .

[0038] Said step 1: the collection volume of the surface water sample is 4.5 L, and it is refrigerate...

Abstract

The invention discloses a method for monitoring the diversity of freshwater benthic animal communities based on an environmental DNA technology. The method comprises the following steps: (1) collecting a surface water sample; (2) filtering a water sample and extracting eDNA; (3) carrying out PCR amplification; (4) performing high-throughput sequencing to obtain a representative sequence of the OTU; (5) establishing a benthonic animal comparison database; and (6) comparing the established benthonic animal database, and performing species annotation on the OTUs representative sequence of the sample to be detected. The method has the advantages of simple sampling, low labor cost, convenient and fast detection, and no need of morphological identification of benthonic animals.

Description

technical field [0001] The invention belongs to the field of molecular ecology, and in particular relates to a method for monitoring the diversity of freshwater benthos communities based on environmental DNA technology. Background technique [0002] Benthic macroinvertebrates, usually referred to as benthic animals for short, refer to the group of aquatic invertebrates living at the bottom of water bodies for all or most of their life history, with a body length of more than 0.5mm. Benthic animals in the water environment are widely distributed and various in variety, mainly including Oligochaeta and Verrucota in the phylum Annelids, Gastropoda and Bivalvia in the phylum Mollusca, Insecta and Crustacea in the phylum Arthropoda, etc. Because benthic animals have the advantages of large individuals, easy collection, fixation and identification, relatively fixed activity places, long life spans, and sensitive responses to environmental changes, they are often used as indicator ...
