Embodiment 1

[0035] Example 1 Extraction, separation and preparation of new phenylpropanoid compounds in yew branches and leaves

[0036] A method for extracting, separating and purifying phenylpropanoids from branches and leaves of Taxus chinensis, comprising the steps of:

[0037] 1) Freshly cut yew branches and leaves, air-dried naturally, pulverized with a pulverizer, and sieved to a 30-60 mesh sieve. Qualified raw materials were added with 60% ethanol aqueous solution at a mass ratio of 1:10, ultrasonically assisted extraction 3 times at a temperature of 50° C., each extraction was 50 min, filtered, and the filtrate was vacuum concentrated to 1 / 3 of the original volume to obtain crude Extract.

[0038] 2) Add 5 times the mass of water to the crude extract, stir, then add petroleum ether with 5 times the mass of the crude extract to extract 5-6 times, and add 5 times the mass of the crude extract to the remaining phase after petroleum ether extraction. Extract with chloroform for 5-6...

Embodiment 2

[0043] Example 2 Antitumor activity test of yew branches and leaves extract and isolated active components

[0044] Preparation of sample solution: Take 5g of the ethyl acetate extract sample of yew branches and leaves, use AB-8 macroporous adsorption resin column to separate, use different concentrations of ethanol for gradient elution, and 20% ethanol eluate is passed through silica gel Column separation, eluting with dichloromethane-methanol system to obtain 100% dichloromethane eluent, after HPLC analysis, enriching and merging fractions with the same components, concentrating under reduced pressure, and freeze-drying to obtain pure components. Sample solution with a concentration of 5 μmol / mL for later use.

[0045] Determination of the tumor activity of the sample: Three cancer cells, human breast cancer (MCF7), human cervical cancer (HELA) and human liver cancer (HepG2), were selected for the anti-tumor activity experiment. Three types of cancer cells in the logarithmi...

Abstract

The invention discloses a taxus chinensis branch and leaf phenylpropanoid compound as well as an extraction and separation method and application thereof, which belong to the technical field of extraction and separation of phenylpropanoid compounds. The new phenylpropanoid compound Pu1 is extracted, separated and prepared from taxus chinensis branches and leaves, and the method is simple to operate, low in production cost and high in yield. The antitumor activity of the extracted and separated phenylpropanoid compound Pu1 is detected by an MTT method, and the antitumor activity is selected from three cancer cells of human breast cancer, human cervical cancer and human liver cancer. Results show that the IC50 values of Pu1 for inhibiting the activity of three cancer cells are 75.7 mumol.L<-1>, 54.7 mumol.L<-1> and 33.8 mumol.L<-1> respectively, and the IC50 values of reference sample etoposide are 4.8 mumol.L<-1>, 7.5 mumol.L<-1> and 5.9 mumol.L<-1> respectively, which indicates that phenylpropanoid compounds Pu1 have certain antineoplastic effects.

Description

technical field [0001] The invention belongs to the technical field of extraction and separation of phenylpropanoid compounds, and in particular relates to a phenylpropanoid compound of yew leaves and its extraction and separation method and application. Background technique [0002] Taxus (taxus), also known as red pine and cypress, is a plant of the genus Taxus in the family Taxaceae. It is a rare and natural anti-cancer plant with a history of 2.5 million years on the earth. There are 11 species of yew in the world, and 4 species and 1 variety in China, namely Chinese yew, northeast yew, Tibetan yew, southern yew, and Yunnan yew. Although the natural yew plants in my country are widely distributed, they are relatively rare in number, rarely able to form forests, and grow relatively slowly. At present, my country's yew resources are mainly cultivated artificially, mainly distributed in Sichuan, Guangxi, Shandong, Heilongjiang and other provinces, and its yew resources acc...
