Embodiment 1

[0055] Embodiment 1, the breeding of novel high-temperature-resistant lactic acid bacteria——Lactobacillus plantarum SG-03

[0056] 1. Preparation of protoplast suspension of Lactobacillus plantarum

[0057] 1. Preparation of activation and bacterial suspension of Lactobacillus plantarum

[0058] The Lactobacillus plantarum used in this example is Lactobacillus plantarum CICC20265, which was purchased from China Industrial Microorganism Culture Collection and Management Center.

[0059] Inoculate the glycerol bacteria of Lactobacillus plantarum CICC 20265 to the slant containing MRS agar medium (18×180mm test tube slant), and cultivate it at 30°C for 24h. This is the F1 generation slant, and then take a ring from the F1 generation slant to The fresh empty slant was cultured at 30°C for 16 hours to obtain the activated Lactobacillus plantarum CICC 20265 strain.

[0060] Then, wash the thalline in the activated slant with sterile water to prepare a bacterium suspension, and the...

Embodiment 2

[0113] Embodiment 2, the fermentation application of Lactobacillus plantarum SG-03

[0114] 1. Fermentation of Lactobacillus plantarum SG-03

[0115] 1. Seed activation and cultivation

[0116] 1.1. Inoculate Lactobacillus plantarum SG-03 glycerol bacteria onto the slant surface of MRS agar medium (18×180mm), and cultivate at 30°C for 48 hours in an incubator.

[0117] 1.2. Wash the conidia with 5ml of sterile saline, transfer to MRS eggplant bottle medium (an eggplant bottle with MRS agar medium), and culture in an incubator at 30°C for 24 hours, which is the activation of the strain Cultivate to obtain the activated Eggplant bottle strain, which is the activated Lactobacillus plantarum SG-03 strain.

[0118] 2. Shake flask seed culture

[0119] Take the Lactobacillus plantarum SG-03 strain activated in step 1, add 20ml of sterile water to elute to prepare the bacterial suspension, transfer it to a 2L shake flask with 500mL of MRS broth medium, and place it in a constant t...

Abstract

The invention discloses a method for preparing a fusion strain. According to the method, lactobacillus plantarum and bacillus licheniformis are used as parent strains, and through fusion of inactivated amphiphilic protoplast and screening aiming at acid production, bacteriostasis and high temperature resistance, the fusion strain is obtained. Experiments prove that the fusion strain which can produce acid and has enzyme production, stress resistance and high temperature resistance of spore bacteria can be prepared by the method for preparing the fusion strain. The lactobacillus plantarum SG-03prepared by the method disclosed by the invention is high in lactic acid yield, has good antibacterial activity, can resist the high temperature, has good processability and is safe to the human andlivestock, free of environmental pollution problems, simple in culture condition, easy to store and suitable for development and application, and living bacteria agent products prepared from the lactobacillus plantarum can promote growth of broiler chickens and improve the productivity of the broiler chickens.

Description

technical field [0001] The invention relates to a strain of lactobacillus plantarum and a preparation method thereof in the field of biotechnology. Background technique [0002] In recent years, the disadvantages caused by the widespread use and even abuse of antibiotics in humans and animals have become increasingly apparent. Long-term use of antibiotics will cause imbalance of normal flora in human and animal intestines, affect the formation of the body's immune function, and even produce multi-drug-resistant "super bacteria" due to long-term abuse of antibiotics. Therefore, finding alternatives to antibiotics has become an urgent task for human survival and development in the future. [0003] Lactobacillus plantarum (Lactobacillus plantarum) is a lactic acid bacterium that has been reported to exist naturally in the intestinal tract of almost all vertebrates and mammals. Lactobacillus plantarum has strong adhesion to the intestinal mucosa and excellent resistance to gas...
