[0040] Example 1 Preparation of canine adenovirus type I penton basal protein subunit vaccine

[0041] (1) Construction of plasmid pColdII-PB: use the genome of CAdV-1 F1301 virus strain as a template, design PB and Knob primers, and use PCR method to amplify PB (1434bp) and Knob (519bp). PCR amplification results are shown in figure 1 .

[0046] After the product was purified, it was digested with SacI and PstI, and the 1434bp fragment was recovered. At the same time, the plasmid pColdII was digested with SacI and PstI, and the 3966 fragment was recovered. The two DNA fragments recovered above were treated with T 4 DNA ligase connection, after the connection product was transformed into competent Escherichia coli DH5α, cultivated on the LB solid medium containing ampicillin, and screened the pColdII-PB and pColdII-Knob containing plasmids (...

Abstract

The invention discloses a canine adenovirus type I subunit vaccine and a preparation method thereof. The invention provides an antigen protein with immunogenicity. The antigen protein is encoded by acanine adenovirus type I penton substrate gene segment, the nucleotide sequence of the encoding gene is shown as SEQ ID No. 1, and the amino acid sequence of the encoded protein is shown as SEQ ID No.2. The antigen protein is high in immunity; high safety, the dog can be effectively protected from the attack of the I-type adenovirus; the antigen protein can be directly used as a canine adenovirustype I subunit vaccine without adding an immunologic adjuvant, the ELISA titer of a specific antibody and the titer of a virus neutralizing antibody in serum of a mouse can be remarkably improved after the antigen protein is injected into the mouse, the proliferation activity of spleen T lymphocytes is improved, and it is proved that the antigen protein has good immunogenicity. After the subunitvaccine disclosed by the invention is used for immunizing foxes for 30 days, the survival rate of the foxes is 100%.

Description

technical field [0001] The invention relates to a canine type I adenovirus subunit vaccine, in particular to a canine type I adenovirus subunit vaccine and a preparation method thereof, belonging to the field of canine type I adenovirus subunit vaccines. Background technique [0002] Canine adenovirus (CAdV) is a double-stranded DNA virus, including type I and type II. Canine adenovirus type 1 (CAdV-1) mainly causes acute septic hepatitis (dogs) and encephalitis (bears and foxes). CAd V-1 is widely distributed all over the world. It is highly pathogenic and has a wide spectrum of infection. In addition to dogs and foxes, CAd V-1 can also cause fatal infections to wild carnivorous mammals (wolves, brown bears, black bears, otters) . [0003] CAdV-1 has strong tolerance, fast transmission speed and wide range of hosts. Vaccine immunization is currently recognized as the most effective way to prevent CAdV-1 infection. The vaccines of canine type I adenovirus mainly include i...
