Problems solved by technology

This method has two outstanding disadvantages: 1) the microspheres have different particle sizes, and must be pre-lyophilized and sieved and separated under aseptic conditions to remove oversized and undersized microspheres. The yield is very low; 2) The drug solution as the inner water phase of the double emulsion inevitably contacts the outer water phase during emulsification and stirring, resulting in drug leakage, and the drug loading of the microspheres decreases
The membrane emulsification method also has a series of disadvantages, which restrict its industrial application: 1) The embryonic microspheres released from the membrane will fuse into large particles when they settle at the bottom of the container, and stirring to prevent their adhesion will cause a certain degree of shearing and quenching. and collision fusion; 2) The gas pressure and the outflow of the polymer solution as the extrusion power are not always linear, and the extrusion rate is first affected by factors such as the concentration, viscosity, drug loading, and room temperature of the polymer solution; 3) Hydrophobicity The gas has considerable solubility in the organic solvent that dissolves the polymer, so that part of the polymer beads float to the water surface after being extruded to form a sheet
[0008] In view of this, for decades, the attempts of microfluidics and membrane emulsification to prepare microspheres have been reported, but they have remained in the research and development stage, and have not become a feasible microsphere production technology so far.

Abstract

The invention discloses a preparation method and a preparation mechanism of embryo microspheres and a preparation method and a preparation device of microspheres. The preparation method comprises thefollowing steps of: conveying a microsphere generation liquid to a porous film positioned in a receiving liquid through a liquid conveying member to form the embryo microspheres; flowing the embryonicmicrospheres falling off the porous film along a channel filled with the receiving liquid to harden the embryo microspheres to form the microspheres; and collecting the microspheres. The flow rate ofthe liquid output by the liquid conveying member is controllable, and the amount of the microsphere generation liquid output per unit time can be directly controlled, so that the particle size and uniformity of the generated microspheres can be better regulated and controlled. In addition, since bubbles are prevented from being mixed into the embryo microspheres, the problem that the microspherescannot be finally formed for the reason that the surface tension distribution of the embryo microspheres changes can be avoided, and the product yield of the microspheres is improved.

Description

technical field [0001] The invention relates to the field of microsphere preparation, in particular to a method for preparing embryo microspheres, a preparation mechanism, a method for preparing microspheres and a preparation device. Background technique [0002] Microspheres are tiny spherical entities with a particle size ranging from 1-250 μm. Polymer microspheres have great potential in the field of medical science because of their good fluidity, ease of injection, and slow release of the contained ingredients. They have been extensively studied since the 1970s . The concept was first proposed from the article "Polymers for sustained release of proteins and other macromolecules" published by R. Langer and J. Folkman in the journal "Nature" (263:793-800). In view of the fact that biological drugs have outstanding curative effect but are difficult to be orally absorbed through the tissue membrane and have to be injected frequently, the author proposes a plan to use degra...
