Embodiment 1

[0044] A preparation method of tissue engineering scar skin model, comprising the following steps:

[0045] S1. Extraction of scar tissue acellular dermal matrix protein:

[0046] S11. Wash the scarred skin twice with PBS containing 10% double antibody, cut the scarred skin into a size of 1cm╳1cm with scissors, and then digest it in neutral protease (2mg / ml) at 4°C overnight;

[0047] S12. separate scar skin dermis and epidermis with tweezers;

[0048] S13. Cut the separated scar skin dermis tissue obtained in step S12 into a size of 2mm╳2mm, and cut the thickness into 2mm, prepare decellularization solution 0.5%SDS+0.5%Triton-100, put the cut scar skin dermis into the prepared decellularization solution, soak overnight at 4°C;

[0049] S14. Take out the soaked scar skin dermis, wash it with sterile PBS for 24 hours and shake it with a horizontal shaker. During the shaking process, change the solution every 2-3 hours. After changing the solution 4-5 times, change the sterile...

Abstract

The invention belongs to the technical field of tissue engineering, and provides a preparation method of a tissue-engineered scar skin model. Scar tissue acellular dermal matrix protein, scar fibroblasts and rat tail collagen are subjected to tissue engineering skin culture so as to obtain tissue-engineered skin. The method comprises the following steps: S1, extracting acellular dermal matrix protein of scar tissue; S2, extracting scar fibroblasts; S3, carrying out mixed culture on the scar tissue acellular dermal matrix protein obtained in the S1, the scar fibroblasts obtained in the S2 and the rat tail collagen; and S4, after one week of culture, adding epidermal stem cells, and performing culturing until the tissue-engineered skin is formed. The technical scheme solves the problem of lack of a proper human scar model in the prior art.

Description

technical field [0001] The invention belongs to the technical field of tissue engineering and relates to a method for preparing a tissue engineering scar skin model. Background technique [0002] Scarring is due to the accumulation of extracellular matrix (ECM) components in the dermis, resulting in structural changes and impaired function of the dermis. After the skin is damaged, the wound coagulation cascade is activated, followed by the recruitment of early inflammatory immune cells, which promote anti-inflammatory and bactericidal wounds, and induce angiogenesis and fibrosis. During this process, fibroblasts are activated and transformed into myofibroblasts, which secrete a large amount of extracellular matrix such as type I collagen, and then transform into scars and promote wound healing. [0003] The composition and structure of ECM play an important role in maintaining the function of tissues. It surrounds cells and is a molecular scaffold and connection channel for...
