The method comprises the following steps of culturing rice seedlings, placing the cultured rice seedlings into a culture pond containing alkali nutrient solution, and meanwhile, adding sodium nitroprusside and pyrazinamide into the culture pond and keeping the concentration of both the sodium nitroprusside and the pyrazinamide at 10-50 mu mol*L<-1>. The method for improving the alkali resistance and the iron content of the rice takes the sodium nitroprusside and the pyrazinamide as exogenous cropgrowth regulator to effectively root growth inhibition and overground part withering which are caused by an alkaline stress environment, significantly improve the alkali resistance of the rice, increase the iron content of the rice and finally increase the biomass and yield of the rice, thereby laying a foundation for research on the adaptive mechanism of the rice under saline-alkali stress.
