Problems solved by technology

[0004] The traditional production mode of long-term vegetative propagation of Wen Yujin is far from being able to meet the market's requirements for the quality and output of Wen Yujin, and cannot adapt to modern production. Therefore, in order to improve the production and quality of Wen Yujin, modern biological cultivation methods must be used. Such as tissue culture technology, in order to obtain the sterilized vaccine quickly and efficiently
At present, there is no detailed report on the sterilized and rapid propagation of yujin. Therefore, this experiment intends to use the sterilized seedlings of yujin to quickly produce yujin through the rapid propagation system of yujin. The implementation provides a theoretical basis, and at the same time provides an effective way for factory production of seedlings to solve the long-term interference with the economic reduction of turmeric growers due to seed retention

Embodiment Construction

[0023] The embodiment of the present invention provides a kind of rapid propagation technology of turmeric sterilized seedlings, comprising the following steps:

[0024] ① Acquisition of explants: cut off the 0.5-1.0cm-long buds from the roots, rinse them under running water, remove surface dirt, carefully peel off the leaf sheaths, and cut them into small sections with stem tips of about 0.5cm. Conducive to the start-up induction of wen yujin;

[0025] ②Sterilization of explants: Move the small section with stem tip to the ultra-clean workbench, sterilize with 75% alcohol for 5-10 seconds, rinse with sterile water for 3-4 times, then place in sterile water, and turn on ozone sterilization After one hour with the ultraviolet sterilizing lamp, filter dry and place in a sterile bottle;

[0026] ③Induced tissue culture seedlings with clustered buds: Inoculate the obtained explanted body temperature turmeric buds into MS medium, add 1.0-4.0mg / L of 6-BA and 0.1-0.5mg / L of NAA to M...

Abstract

The invention provides a rapid propagation technology for aseptic seedlings of curcuma wenyujin. The rapid propagation technology comprises the following steps: (1) explants are obtained; (2) the explants are sterilized; (3) cluster buds are induced into tissue culture seedlings; (4) root tips are induced for producing calluses; (5) the calluses are induced into the aseptic seedlings of the curcuma wenyujin; and (6) the aseptic seedlings of the curcuma wenyujin propagate. The invention adopts the mean of tissue culture seedling for inducing root tip meristem to produce the calluses and obtainsthe aseptic seedlings after differentiation , thereby effectively solving the difficult problem that aseptic meristem is difficult to get directly from the explants, ensuring the obtainment of the aseptic seedlings and solving the problem that the tissue culture seedlings produced through the induction to the explants cannot continuously propagate due to carried endophyte. The technology providedby the invention realizes the continuous propagation of the aseptic seedlings in a MS culture medium added with 2.0 mg / L of 6-benzylaminopurine(6-BA) and 0.5 mg / L of naphthlcetic acid(NAA) and fulfills propagation and rooting synchronously, thereby omitting the processes of rooting and seedling development and reducing the cost in tissue culture.

Description

technical field [0001] The invention relates to the field of warm turmeric propagation technology, in particular to a rapid propagation technology of warm curcuma sterilized seedlings. Background technique [0002] Wenyujin is a perennial herb of the Zingiberaceae Curcuma genus. It has the value of medicine, health care, food, beauty and garden ornamental. It is an important economic crop. The part of Wenyujin used as a traditional Chinese medicine is a tuber, which needs to be processed differently to form "Wenyujin", "Wenyujin", and "Piece Turmeric". It is often used to relieve qi stasis, calm pain, and fight inflammation. The volatile oil containing sesquiterpene alcohols and sesquiterpene compounds is extracted from the rhizome of turmeric. The ingredients in it can effectively treat tumors and reduce the course of thrombus. It is a very effective drug and has a very broad development prospect. At the same time, Wen Yujin is also often used to reduce the degree of plate...
