Problems solved by technology

Consumers are prone to enteral diseases such as dysentery if they eat food with serious microbial excess, which may cause symptoms such as vomiting and diarrhea, endangering human health and safety
[0003] Colony counting is one of the most basic and important tasks in routine microbial inspection. At present, manual counting of culture medium is mainly used. However, in the process of colony culture and manual counting, the following factors will cause errors in accurate quantification
[0004] Spreading growth and pollution directly hinder the counting or inhibit the growth of bacteria in the specimen, affecting the accuracy of the counting results
Different uniformity of cultured bacteria blocks will cause uneven growth of colonies, and different degrees of dispersion will cause colonies to grow in chains to a certain extent, resulting in difficulty in counting
Particulate impurities in the sample cause counting errors. Some solid particles in the sample are very similar in size and shape to bacterial colonies, which are difficult to distinguish in shape, resulting in counting errors
Colony growth is too slow, resulting in long culture counting time required

Abstract

The invention discloses a detection device and method for colony growth states based on the multi-mode imaging technology. The device includes a white light source, a coherent laser source and a fluorescent excitation light source, which are used as three kinds of light sources to irradiate the colony samples above the slope for sample image acquisition; a color camera, a high-speed camera and a cooling camera, which are used as three kinds of cameras to collect the images of colony samples irradiated by three kinds of light sources above the sample respectively; an imaging system, including imaging lens, a spectroscope, a filter and a relay eyepiece, which is set between the colony samples and the cameras to realize the corresponding imaging between the colony samples irradiated by threekinds of light sources and the three kinds of cameras, and the imaging parameters are identical; and a computer, through which the imaging signals are received to complete image fusion processing. Multimodal optical imaging and information fusion method are used to eliminate the background effect of culture medium, improving the accuracy of colony counting, and realizing all-round detection and discrimination of colony growth status.

Description

technical field [0001] The invention relates to the field of optical detection, in particular to a multimodal imaging technology-based detection device and method for colony growth state. Background technique [0002] In the field of food and drug safety, microorganisms are the main cause of food spoilage, microbial toxins (pathogenic bacteria, parasites) and the prevalence of infectious diseases (hepatitis virus), and the food-borne diseases caused by them are the number one problem of food safety in my country . In my country's food hygiene management, microbial indicators are divided into the total number of colonies (total number of bacteria), coliforms, molds, yeasts and pathogenic bacteria. Colonies are mainly used as a sign to determine the degree of food contamination by microorganisms. On the one hand, the number of colonies can be observed as a sign of the degree of food contamination. This is also included in the national standard GB 4789.2-2010 "National Food Saf...
