Problems solved by technology

At present, the internationally recognized first-line treatment strategy for RA is the early combined use of antirheumatic drugs to improve the condition, control the onset of RA, and reduce the disability rate, but about 30% of patients still cannot be relieved, and uncontrolled active RA can cause joint pain Damage, disability, reduced quality of life, cardiovascular disease and other comorbidities, which can also increase patient anxiety and make patients have to endure emotional depression and a huge financial burden

Embodiment Construction

[0055] The following will clearly and completely describe the technical solutions in the embodiments of the present invention with reference to the accompanying drawings in the embodiments of the present invention. Obviously, the described embodiments are only some, not all, embodiments of the present invention. Based on the embodiments of the present invention, all other embodiments obtained by persons of ordinary skill in the art without making creative efforts belong to the protection scope of the present invention.

[0056] Primers for detecting inflammatory factors in rheumatoid arthritis rats and a method thereof, comprising the following steps:

[0057] Step 1: Prepare test animals and their tissues

[0058] Select 24 Wistar female rats with good health and similar weight (170 ± 10g) at the age of 42 ± 2 days, and according to the principle of similar average body weight between each group, they are randomly divided into 2 test groups: Group I, blank control group (n = ...

Abstract

The invention relates to the field of biomedical technology, a primer for detecting inflammatory cytokines in rats with rheumatoid arthritis and a method thereof are disclosed, when the test animals and their tissues are ready, total RNA was extracted from CIA rat joints, the purity and concentration of total RNA were detected, and 5 pairs of specific primers were synthesized by reverse transcription of total RNA. Using CIA rat cDNA as template, 15 [mu]L system PCR was used to detect total RNA. In the gel imaging system, the ultraviolet ray was separated by a partition plate, and the rat RelA,TLR4, AKT1, IL6, NF-kB cDNA fragment gel, rapid agarose gel DNA recovery kit, and ligation of the recovered DNA fragment to T-Vector, screening and sequencing of transformed blue and white spots. A CIA rat model is established, detecting the expression of NF-kB subunit p65, toll-like receptor 4, AKT serine / threonine kinase 1, interleukin 6 and NF-kB subunit in the joints of rats with rheumatoid arthritis by molecular biological methods is of great significance for the early prevention and treatment of RA. The results showed that the expression of NF-kB subunit p65, toll-like receptor 4, AKT serine / threonine kinase 1, interleukin 6 and NF-kB subunit in the joints of rats with rheumatoid arthritis was significantly higher than the of normal joints.

Description

technical field [0001] The invention relates to the technical field of biomedicine, in particular to a primer for detecting inflammatory factors in rheumatoid arthritis rats and a method thereof. Background technique [0002] Rheumatoid arthritis (RA) is a common inflammatory disease of the immune system, affecting approximately 1% of the population worldwide. RA manifests as persistent synovitis, systemic inflammation, and continuous production of autoantibodies. The exact etiology of RA is unclear, but environmental and genetic factors are involved in the pathogenesis of RA. At present, the internationally recognized first-line treatment strategy for RA is the early combined use of antirheumatic drugs to improve the condition, control the onset of RA, and reduce the disability rate, but about 30% of patients still cannot be relieved, and uncontrolled active RA can cause joint pain. Damage, disability, reduced quality of life, cardiovascular disease, and other comorbiditi...
