Abstract
The invention provides a vaccine strain rSHA-△200, its construction method and application, and relates to the field of influenza vaccines. The vaccine strain rSHA-Δ200 includes an HA gene that does not contain an NRT glycosylation site. The present invention finds out potential glycosylation sites on the HA protein through analysis, and deletes glycosylation sites through site-directed mutagenesis technology to construct HA expression plasmids with deletion of multiple glycosylation sites, and utilizes 8 plasmids to reverse The recombinant vaccine strain was obtained by genetic technology, and the antiserum was prepared. Through comparison, it was found that the rSHA-△200 obtained by the present invention has good cross-immunity and serum cross-neutralization ability, and is an ideal H9 subtype AIV inactivated vaccine candidate strain.
Description
technical field [0001] The invention relates to the technical field of influenza vaccines, in particular to a vaccine strain rSHA-Δ200 and its construction method and application. Background technique [0002] H9 subtype AIV is usually low-pathogenic avian influenza, which causes low poultry mortality. However, due to its wide distribution and fast transmission speed, the egg production of laying hens will drop sharply after infection, and broiler chickens will grow slowly. Reducing the feed-to-meat ratio has caused huge economic losses to the poultry farming industry. H9 subtype AIV infection is likely to lead to a sharp decline in the poultry's own immunity, resulting in concurrent or secondary infections of other bacteria or viruses leading to increased mortality. The H9 subtype AIV will also serve as the donor of the internal genes of other subtype influenza viruses, providing conditions for the generation of new influenza virus strains, triggering the outbreak of new i...
