Problems solved by technology

The identification of clematis in different cultivation methods on the market is limited to empirical judgment, but the difference between 3-month-old bottle seedlings, 4-month-old bottle seedlings, or 6-month-old seedlings planted in the soil is not significant, leading some unscrupulous merchants to directly sell Tissue-cultured bottle seedlings were put on the market, shoddy as good ones, causing confusion in the sales market, and seriously hindering the healthy development and scale expansion of the clematis industry

Embodiment 1

[0028] Embodiment 1 Detection method of the present invention

[0029] (1) Sample treatment: wash the fresh samples to be tested in batches for 3 to 5 times until the water becomes clear and there is no sediment on the surface of the samples, place them in a 60°C oven to dry to constant weight, crush them through a 60-mesh sieve, and take an appropriate amount for each batch. Put it into the finger tube and tamp it so that the sample height is between 2cm, to be tested;

[0030] (2) Spectrum acquisition and preprocessing: Scan the sample with NIRFlex N-500 near-infrared spectrum analyzer, acquisition parameters: scanning range is 5000-10000cm -1 , the number of scans is 32, and the resolution is 8cm -1 , each sample is reloaded and scanned 3 times. Before each scan, the sample in the finger tube should be tamped to make the sample even and flat. The system will automatically store the NIRS spectrum of the scanned sample into the spectral database; in the NIRCal 5.4 data proce...

[0036] Embodiment 2 detection method of the present invention

[0037] (1) Sample treatment: wash the fresh samples to be tested in batches for 3 to 5 times until the water becomes clear and the surface of the samples is free of sediment, put them in a 55°C oven and dry them to constant weight, crush them through a 100-mesh sieve, and take an appropriate amount for each batch. Put it into the vial and tamp it down so that the height of the sample is between 1cm, to be tested;

Abstract

The invention discloses an identification method for anoectochilus roxburghii with different cultivation methods; a near infrared spectroscopy analysis technology is adopted for identification, wherein the method includes the following steps: (1) taking a to-be-detected sample powder, loading the sample, and collecting a near infrared spectrogram; and (2) adopting a PCA-Cluster clustering analysismethod as a calculation method, selecting unit length normalization (nle) and first-order smooth derivation (dgl) pretreatment for pretreating the near infrared spectrogram, and identifying the anoectochilus roxburghii with different cultivation methods by score values of sample main components. The identification method has the advantages of being simple, fast, accurate, green and environmentally friendly, has no damage to the sample, can effectively identify 3-month bottle seedlings, 4-month bottle seedlings or 6-month in-soil planted seedlings of anoectochilus roxburghii, and has good application prospect.

Description

technical field [0001] The invention relates to a method for identifying clematis in different cultivation modes. Background technique [0002] Anoectochilus roxburghii is a plant of the genus Orchidaceae, mainly distributed in Fujian and Taiwan regions of my country, and is a rare and endangered perennial medicinal material. Auranthus clematis has the effect of clearing away heat and cooling blood, dispelling wind and dampness, and it is mainly used to treat diseases such as nephritis, cystitis, diabetes, bronchitis, and convulsions in children. Clematis clematis has high medicinal and health care value, but wild resources are scarce, and currently the market is dominated by cultivated products. The cultivation methods are mainly divided into tissue culture and planting in the soil. Most of the tissue culture products are tissue culture bottle seedlings of 3 to 5 months old, and most of the soil planting products are 3 to 5 month old bottle seedlings transplanted into the ...
