Problems solved by technology

Commercial lipid droplet fluorescent dyes mainly include NileRed (NileRed) and BODIPY493 / 503, but they still have some important defects: strong fluorescent background and small Stokes shift
[0003] However, these AIE lipid droplet fluorescent probes still have the problem of short excitation wavelength, resulting in strong background fluorescence and low penetration depth in tissue sections.
To solve these problems, a lot of effort has been devoted to the synthesis of fluorescent dyes excited by long wavelengths, but there are few successful examples
The main challenge of this scheme is that increasing the intermolecular conjugation causes synthesis difficulties, the molecular weight becomes larger, the molecular hydrophobicity increases, the cell penetration decreases, and the quantum yield of red light is low.

Abstract

The invention provides a fluorescent probe and its application. The probe has a donor-π-acceptor electronic structure, has twisted intramolecular charge transfer and aggregation-induced luminescence properties, and can be excited by two-photons. It overcomes the self-absorption problem caused by the large background and small Stokes shift of commercial lipid droplet fluorescent dyes, and has good biocompatibility, high brightness, low background and good photostability. At the same time, the present invention utilizes the two-photon excitation process, which can reduce autofluorescence, increase signal-to-noise ratio, improve three-dimensional resolution and photostability; the probe proposed by the present invention can be applied to lipid droplet imaging of various cells and tissue slices.

Description

technical field [0001] The present invention relates to two-photon fluorescent probes with aggregation-induced luminescent characteristics and their application in lipid droplet imaging in biological cells. Background technique [0002] Lipid droplets (Lipid droplets, LDs) are composed of phospholipid single-molecule surface layer and neutral lipid inner core, which are the main storage places of neutral lipids in cells, and widely exist in a variety of animal and plant cells. Recent studies have found that lipid droplets are not an "inert" energy storage, but an active multifunctional organelle, and its abnormalities are associated with diseases such as obesity, type II diabetes, fatty liver, hyperlipidemia and atherosclerosis are closely related. Therefore, the detection of lipid droplets is of great significance to biomedical research and clinical diagnosis. In recent years, fluorescence detection method has gradually become an important research method in biomedical re...
