The preparation method comprises the following steps of after collecting rumen liquor, performing direct culture or in vitro culture so as to prepare strain liquor; crushing fine and coarse feed products, performing pulping to obtain pulp, and pumping the pulp into a fermenter; after raising the temperature and introducing CO2 for deoxygenization, adding trace element nutrient buffering liquor and the strain liquor, strictly performing anaerobic fermentation to prepare fermentation liquor, then performing solid-liquid separation to obtain a solid wet base and liquid nutrient liquor; and directly concentrating the liquid nutrient liquor to prepare concentrated liquid nutrient liquor; mixing the fermentation liquor solid wet base with feed raw materials, pure water, compound strain liquor and compound enzymes, and performing enzymolysis fermentation, so as toprepare fermented and enzymatically hydrolysed solid wet bases; mixing the fermented and enzymatically hydrolysed solid wet bases with the concentrated nutrient liquor to prepare the fermentation feedrich in volatile fatty acid calcium.
