Problems solved by technology

Fhb1 has been proven to be the most resistant and stable head blight resistance gene. The current limiting factor for marker-assisted selection of Fhb1 is the lack of low-cost diagnostic markers, although its linkage markers Xgwm493, Xgwm533, UMN10 and Xsnp3BS-8, etc. There are certain applications in breeding, but they are greatly affected by the genetic background

Embodiment 1

[0041] Embodiment 1, the method for the acquisition of molecular marker PFT-CAPS and identification of wheat head blight resistance gene PFT

[0042] 1. Acquisition of three alleles and polymorphic sites of PFT

[0043] 1. Design of specific primers for PFT amplification

[0044]Using the sequence of the PFT coding region in the Fhb1 segment of Sumai No. 3 (GenBank accession number: KX907434) as a probe, sequence alignment was carried out in the EnsemblPlants database (http: / / plants.ensembl.org / index.html) to obtain each Gene homologous sequence, using the obtained homologous sequence and the corresponding gene sequence of Sumai No. 3 as a reference, design specific primers for amplifying the open reading frame (ORF) 3B chromosome of the gene. The primer sequences are shown in Table 1. Primer binding positions such as figure 1 shown.

Abstract

The invention discloses a mark for detection or auxiliary detection of wheat FHB (Fusarium Head Blight)-resistant genes PFT (Pore-Forming Toxin-Like) and a use method thereof. 204 parts of PFT genes in different varieties are subjected to PCR amplification and sequencing; the condition that 14 nucleotide differences totally exist between PFT-I allelic genes and PFT-II allelic genes in three kinds of allelic genes of the gene is discovered; and a diagnostic mark PFT-CAPS of the PFT genes is developed according to the 1471rd site deoxyribonucleotide polymorphism of the FPT genes. Experiments prove that the molecular marker PFT-CAPS can be used for the identification of the wheat FHB-resistant genes PFT and the auxiliary breeding of the wheat FHB-resistant molecular markers.

Description

technical field [0001] The invention belongs to the field of biotechnology, and in particular relates to a marker for detecting wheat head blight resistance gene PFT and a method for using it. Background technique [0002] Fusarium head blight (FHB) caused by Fusarium graminearum is a widespread fungal disease that seriously affects wheat yield and quality. The middle and lower reaches of the Yangtze River in my country and the Northeast wheat region are its frequent and re-occurring areas. In recent years, affected by global climate change and the popularity of straw returning under the wheat-corn rotation system, the disease has shown a trend of expansion and aggravation, and has become a common disease in the Huanghuai wheat region. At present, the average annual occurrence area of wheat scab in my country exceeds 5.333 million hm 2 , of which 2012, 2015 and 2016 were particularly serious. For example, the average annual occurrence area in Jiangsu Province from 2012 ...
