Abstract

The invention discloses a paraffin section method for pomegranate seed particles. The paraffin section method comprises the following steps: (1) material selection and fixing; (2) dehydration and transparency: carrying out gradient dehydration, carrying out treatment for 2 h in each gradient, and then converting into absolute ethyl alcohol treatment for 1.5 h twice; adding a mixed solution of absolute ethyl alcohol and xylene in a volume ratio of 2: 1 and a mixed solution of absolute ethyl alcohol and xylene in a volume ratio of 1: 2 for 2 h, and then converting into pure xylene treatment for 1.5 h twice; (3) waxing and embedding: putting the transparent materials into a mixed solution of xylene and paraffin in a volume ratio of 1: 1 for waxing for 4 to 5 h, then putting in a mixed solution of xylene and paraffin in a volume ratio of 1: 2 for waxing for 4 to 5 h, then waxing for 4 to 5 h with pure paraffin, and then continuously waxing for 3 to 4 d by changing the pure paraffin; (4) section and spreading; and (5) dyeing to be transparent, and mounting.
