Embodiment 1

[0107] This embodiment provides a gadolinium polytungstate composite nanomaterial, which is composed of gadolinium polytungstate, chitosan and small interfering RNA.

[0108] Preparation:

[0109] 1) Gadolinium polytungstate crystal (named GdW 10 )Synthesis

[0110] In a typical experiment, first weigh 8.3g Na 2 WO 4 2H 2 O was dissolved in 20mL ultrapure water to form a uniform and transparent aqueous solution, and then under the action of magnetic stirring, acetic acid was slowly dropped into the above solution to adjust the pH value of the above solution to 7.40-7.50. Then at room temperature and under the action of magnetic stirring, 2mL concentration was 477.95g mol -1 GdCl 6 ·6H 2 O was added dropwise to the above solution, and then the whole solution system was placed on a heating platform and heated to 85 °C. After the solution became completely clear and transparent, it was allowed to cool to room temperature, and then crystals were precipitated. In order to m...

Abstract

The invention provides a purpose of a gadolinium polytungstate-containing nanometer material as a sensitizing agent. The gadolinium polytungstate-containing nanometer material has the purposes that redox reaction can be carried out on gadolinium polytungstate and glutathione, so that the content of the glutathione is lowered, and internal sensibilization is realized. The invention also provides a gadolinium polytungstate composite nanometer material which is prepared from the gadolinium polytungstate, chitosan and small interfering RNA. The gadolinium polytungstate composite nanometer material has good radiation-sensitizing performance, can absorb a great quantity of reactive oxygen species (ROS) generated by X rays, can also lower expression of HIF-1 alpha at a tumor site and inhibits self repair of DNA with double-stranded breaks; and in addition, most importantly, redox reaction can be carried out on gadolinium polytungstate and glutathione (GSH) in vivo, so that the content of the glutathione in vivo is lowered, more effective reactive oxygen species are generated, effective combination of internal sensibilization and external sensibilization is realized at the same time, and an ideal radiation-sensitizing effect is achieved.

Description

technical field [0001] The invention belongs to the technical field of nanomaterials and radiosensitizing drugs, and relates to the use of a nanomaterial containing gadolinium polytungstate as a sensitizer, in particular to a chitosan-modified gadolinium polytungstate composite nanomaterial. Use as a sensitizer. Background technique [0002] Radiation therapy (RT) is one of the most widely used clinical methods for treating cancer. Its main principle is to use high-intensity ionizing radiation (X-ray or γ-ray) with very strong penetrating power to inhibit tumor growth. During radiotherapy, ionizing radiation interacts with water or oxygen inside the tissue to generate a large amount of cytotoxic reactive oxygen species (ROS) to induce damage to the double strands of cellular DNA. Therefore, in order to enhance ionizing radiation-induced cell damage during radiotherapy, the generation of sufficient reactive oxygen species is necessary to cause DNA double-strand damage by rea...
