Problems solved by technology

[0008] There are no relevant reports at home and abroad on the effect of kaempferol on retinal pigment epithelial cells under oxidative stress. Whether kaempferol may delay the occurrence and development of AMD by reducing the oxidative stress damage of retinal pigment epithelial cells remains to be seen. Research

[0035] (1) H 2 o 2 Induced ARPE-19 injury model of retinal pigment epithelial cells (H 2 o 2 model group)

[0036] ARPE-19 cells were subcultured according to 2×10 5 cell / ml drop plate, 100ul / well. After 24h, different concentrations of H 2 o 2 (0mM, 0.5mM, 1mM, 1.5mM, 2mM) acted on ARPE-19 cells for 24h respectively. Coloring: Add 20ul of MTT solution to each well and continue to incubate for 2-4h. Colorimetry: Select a wavelength of 490nm, measure the light absorption value of each well on an enzyme-linked immunosorbent monitor, record the results, and use different concentrations of H 2 o 2 The cell growth curve is drawn as the abscissa and the absorbance value as the ordinate. When the cell viability was around 50%, the H 2 o 2 The concentration was used as the modeling concentration of the oxidative damage model.

[0037] Depend on Figure 2A It can be seen that when H 2 o 2 The cell survival rate is 57.4% when the concentration is 1mM, so 1mM H 2 o 2 De...

Abstract

The invention discloses application of kaempferol to preparation of a medicament for preventing and treating retina injury diseases. The kaempferol exists in a plurality of edible plants (including fruits, vegetables and beans) and traditional Chinese medicine (chrysanthemum, raspberry, semen cuscutae, radix astragali seu hedysari and ginkgo leaf). The protective effect of the kaempferol on human retinal pigment epithelial cells (ARPE-19) suffering from oxidative stress injury induced by hydrogen peroxide (H2O2) is observed, and the expression change of molecules of the kaempferol relevant to apoptosis after the retinal pigment epithelial cells are treated by H2O2 is observed in order to discuss the anti-apoptotic effect of the kaempferol on the retinal pigment epithelial cells suffering from oxidative injury and the molecular mechanism of the kaempferol, so that a theoretical basis is laid for the prevention and treatment of eye diseases relevant to retinal pigment epithelial cell injury through the kaempferol.

Description

technical field [0001] The present invention relates to a new pharmacological effect of a natural plant monomer, in particular to the application of kaempferol in the preparation of health products or new drugs for the prevention and treatment of retinal damage diseases, and to develop new methods for the prevention and clinical treatment of ophthalmology-related diseases and ideas. Background technique [0002] The retinal pigment epithelium (RPE) has complex biological functions, including providing nutrients to the outer cells of the sensory retina, phagocytosis and digestion of the outer segmental membrane of photoreceptor cells, and maintaining metabolism. RPE adheres very closely to the glass membrane (Bruch membrane) in the innermost layer of the choroid, and forms a unified functional unit together with the choriocapillary layer, that is, the RPE-glass membrane-choriocapillary complex, which is important for maintaining the microenvironment of photoreceptors effect....
