Embodiment 1

[0013] A method for preparing a sperm electron microscope specimen, comprising the following steps:

[0014] a. Asphyxiate the experimental mice by means of experiments, soak the experimental mice in 75% alcohol for sterilization, and cut a V-shaped incision in the abdomen of the experimental mice to expose the epididymis; separate the epididymis, and remove the epididymis from the experimental mice Take out the abdomen and wash it with PBS until the liquid is clear and there is no blood residue. Cut the epididymis short, keep the tail of the epididymis, and soak it in 2.5% glutaraldehyde for 2 hours;

[0015] b. Place the cauda epididymis treated in step a under the environment of 5° C. for 26 hours, and trim it into a strip-shaped electron microscope specimen with a size of 1 mm×1 mm×2 mm for electron microscope observation.

Abstract

The invention discloses a sperm electron microscopic sample slide preparation method in the field of microscope slides preparation. The method comprises the following steps: a, killing a laboratory rat, sterilizing by immersing the laboratory rat in 75% alcohol, cutting the laboratory rat in the abdomen to expose epididymis; separating epididymis, taking the epididymis out of the laboratory rat's abdomen, washing with PBS liquor until the liquor is clear and there is no residual blood, cutting the epididymis short, retaining tail of epididymis, and immersing (fixing) the tail of epididymis in 2.5% glutaraldehyde; and b, placing the processed tail of epididymis in an environment of 3-5 DEG C for 20-26h, and making the electron microscopic sample. According to the scheme, the sperm is avoided from being directly contacted with any reagent used in the slide preparation process. Thus, the sperm is prevented from being dispersed by the reagent, success rate of the slide preparation is raised, wastes of various raw materials used in the slide preparation process are avoided, and work efficiency is also enhanced.

Description

technical field [0001] The invention relates to the field of preparation of microscope slides, in particular to a method for preparing sperm electron microscope specimens. Background technique [0002] Due to the weak penetrating ability of the electron beam generated by the electron microscope, it is necessary to cut the specimen into thin slices with a thickness of less than 0.1 μm. The ultrathin section technique is the most commonly used and basic preparation technique for preparing ultrathin sections. The production process basically needs to go through steps such as material collection, fixation, dehydration, soaking, embedding polymerization, sectioning and staining. [0003] In laboratory experiments, in order to reduce the interference of uncontrollable factors, the observation target is usually processed in a targeted manner to avoid inclusion of other interference factors. For example, in the experimental study of male reproductive ability, it is often necessary...
