Problems solved by technology

Guo, Z. etal, Chem. Commun., 2007, 2491–2493 reported a hydrophilic stationary phase of maltose modified on the surface of silicon spheres. This method uses "click chemistry" to fix maltose containing azide groups to On the surface of silicon spheres, the immobilization rate of maltose is effectively improved, and its ability to separate polar substances is significantly improved. However, this technology belongs to the metal-catalyzed reaction between azide and alkyne, and the material only modifies the sugar. hydroxyl

Embodiment 1

[0019] 1. Preparation of monodisperse core-shell polymer nanoparticles

[0020] In a 150mL round bottom flask, add 80mL of toluene, add 6g of silica gel particles with a particle size of 5μm, and 3g of 3-mercaptopropyltriethoxysilane, and sonicate for 1 minute, so that the added reagents and particles are evenly dispersed in the solution In the process, a condenser was connected to the flask, a magnetic stirrer was added, and the magnetic stirrer was kept at a speed of 300 rad / min. The reaction device was placed in an oil bath, heated to reflux for 6h, then the reaction was stopped and cooled to room temperature. Then use a high-speed centrifuge to centrifuge at a speed of 10,000 rad / min, remove the supernatant, use toluene, acetone, methanol, and acetone to filter and wash in sequence, repeat the suction and filter washing three times, and vacuum dry in a vacuum oven at 50°C for 24 hours. Hours, the modified silica gel particles of the silylating agent were obtained.

Abstract

The invention relates to the field of polymer material and analysis, and relates to organic-inorganic hybrid core-shell-structured microsphere particles, and a preparation and an application thereof. According to the invention, inorganic silica gel particles are adopted as cores, and are modified with a silanization agent; with a thiol-ene click chemistry technology, 3-allyloxy-2-hydroxy-1-propanesulfonic acid and methylene diacrylamide are wrapped on the surfaces of the cores, such that the organic-inorganic hybrid core-shell-structured nano / micro particles with smooth surface and with hydrophilic functional groups such as hydroxyl group, sulfonic acid group, amide and the like are formed. According to the invention, the hydrophilic functional groups such as hydroxyl group, sulfonic acid group, amide and the like are introduced to the surface of the material through the thiol-ene method, such that the defects of complicated steps and low reaction efficiency of a traditional post-modification method are solved, and a polymer three-dimensional-structured hydrophilic layer is formed. Better advantage is shown when the particles are used in hydrophilic chromatography. The microsphere particles can be used in hydrophilic interaction chromatography for separating and enriching glycopeptide, and have good practical value and application prospect in the fields of separation analysis and sugar proteomics.

Description

technical field [0001] The invention relates to an organic-inorganic hybrid microsphere particle and its preparation and application, in particular to an organic-inorganic hybrid core-shell structure microsphere particle with hydroxyl, sulfonic acid, amide and other functional groups at the end. Background technique [0002] Strongly polar compounds (such as oligosaccharides, glycosides, glycopeptides, etc.) play an important role in drugs and organisms, so their separation and analysis have gradually become the focus of attention. However, reversed-phase high-performance liquid chromatography (RP-HPLC), the most widely used high-efficiency separation technique, has poor retention for highly polar compounds. Hydrophilic Interaction Liquid Chromatography (HILIC) as a chromatographic technique for the separation of highly polar compounds (Strege, M.A.etal, Anal.Chem.2000, 72, 4629-4633; Wang, X.D.etal, J.Chromatogr.A , 2005, 1083, 58-62) have been widely developed in recent y...
