Embodiment 1

[0043] Patient Li XX, the examination of the cervix found that the surface of the cervix was smooth or erosive, hard, and easy to bleed when touched.

[0044] Step (1). Pretreatment of exfoliated cells:

[0045] The gynecologist used a special cervical brush for gynecology to obtain exfoliated cells from the patient's cervix, put them into 10-50wt% methanol solution to fix the exfoliated cells, and obtained the pretreated sample solution, which was stored at 0°C;

[0046] Step (2). Centrifugal aggregation of exfoliated cells:

[0047] First add 20μl 1wt﹪saponin to the test tube, then add 5ml of the sample solution obtained in step (1), centrifuge at 2000 rpm for 10 minutes, and discard the supernatant;

[0048] The saponin is used to destroy red blood cells in the sample solution;

[0049] Step (3). The total amount of exfoliated cells is counted:

[0050] Step (2) The exfoliated cell pellet after centrifugation is collected in a test tube, and 2ml of phosphate buffer solutio...

Abstract

The invention discloses a squamous cell carcinoma SCC-TCT combined detection method. The squamous cell carcinoma SCC-TCT combined detection method comprises the following steps: sampling exfoliated cells at a cervical part, and preprocessing to obtain a sample solution; centrifugating saponin and the sample solution, taking precipitate, adding a phosphate buffer solution, uniformly mixing, and counting the total cell number; centrifugating a counted cell suspension, adding the phosphate buffer solution for freezing and thawing at least once to obtain a sample solution of which an SCC antigen is dissociated and released; performing intracellular SCC antigen enzyme-labeled determination analysis; carrying out judging and screening according to a determination result, screening out a sample higher than a critical value, and preparing the sample solution corresponding to the sample into a uniform slide on a liquid-based cytometer, fixing, performing papanicolaou staining, performing morphological cell analysis by using a microscope, and judging the morphology of a tumor cell. According to the squamous cell carcinoma SCC-TCT combined detection method, by an SCC-TCT combined detection technology, 80-90% of normal samples are screened out, so that the working efficiency of a pathological doctor can be improved.

Description

technical field [0001] The invention belongs to the field of biotechnology, and relates to a method for joint detection of human squamous epithelial cancer antigen cells (SCC) and liquid-based cells (TCT), in particular to a human exfoliated cell, that is, exfoliated squamous epithelial cells Cancer antigen (SCC) detection is a method to screen out positive or suspected samples and then conduct liquid-based cell (TCT) pathological cell morphology analysis. Background technique [0002] Cervical cancer is one of the most common malignant tumors of the female reproductive tract, and its incidence rate is the second among female malignant tumors, accounting for about 11% of all tumors. In some developing countries, the incidence of cervical cancer even exceeds the incidence of breast cancer, becoming the first female malignant tumor. [0003] Squamous cell carcinoma antigen (SCC) is a tumor marker with good specificity and the earliest diagnosis of squamous cell carcinoma. It...
