Problems solved by technology

The attenuated live vaccine is safe for SPF chickens, but the immune protection effect of chickens with maternal antibodies is not ideal
Although the immune protection of moderately virulent and highly virulent vaccines is much higher, they will damage the bursa of Fabricius due to their certain pathogenicity.
At the same time, due to the interference of maternal antibodies and the characteristics of IBDV, traditional vaccines can form an inevitable immune blank period in practical applications.

Embodiment Construction

[0019] The technical solution of the present invention will be described in further detail below. It should be noted that the various embodiments of the present invention can be combined in any way as desired.

[0020] The first aspect of the present invention provides a recombinant turkey herpesvirus (HVT) expressing the VP2 protein of infectious bursal virus, specifically, the gene encoding the VP2 protein is inserted into the non-replication region of the HVT genome.

[0021] In a specific embodiment, the non-essential region for replication can be, but not limited to, HVT genome HVT053 and HVT054, as long as this region is not essential for genome replication, it can be used as an insertion region.

[0022] Since it has been proved by experiments that the expression level of VP2 on the herpesvirus of turkey (HVT) vector is positively correlated with the protection of avian bursal disease (IBD) [7]. Therefore, in a specific embodiment of the present invention, the gene enc...

Abstract

The invention provides a recombinant turkey herpesvirus vaccine expressing infectious bursal disease virus VP2 protein and application thereof. According to the invention, on the basis of a Fosmid library continuously covering the whole genome of herpesvirus of turkey (HVT), a VP2 gene carrying a Pec composite promoter is inserted into site HVT053 and site HVT054 in the nonessential region of HVT replication by using Red ET homologous recombination technology and Gateway LR clone technology so as to obtain recombinant HVT (rHVT-VP2). The recombinant HVT (rHVT-VP2) can provide a complete protecting force on the infectious bursal disease virus and is applicable to prevention of the infectious bursal disease virus.

Description

technical field [0001] The invention belongs to the field of molecular biology and biotechnology, in particular to a turkey herpes virus vaccine recombined with infectious bursal virus VP2 protein and its application in preventing infectious bursal disease. Background technique [0002] Infectious bursal disease (IBD) is an acute, highly contact immunosuppressive infectious disease of poultry caused by chicken infectious bursal virus (IBDV). , immunosuppression and high chick mortality cause huge economic losses to poultry farming [1]. Currently commercialized live virus vaccines for infectious bursal disease are divided into three categories: attenuated, poisoned and highly virulent. The attenuated live vaccine is safe for SPF chickens, but it is not ideal for the immune protection of chickens with maternal antibodies. Although the immune protection of moderately virulent and highly virulent vaccines is much higher, they can damage the bursa of Fabricius due to their cert...
