The method comprises the following steps: getting a urine sample, carrying out enzymolysis in constant-temperature water bath, extracting androsterone, removing 13C which is labeled on C3 in an androsterone molecule to obtain ANAD; and dissolving ANAD by tert-Butyl methyl ethe, and analyzing in a isotope ratio mass spectrometer, thereby obtaining the delta value of ANAD, wherein if the absolute value of the difference between the delta value of androsterone and the delta value of ANAD is greater than 1.2 times delta unit value, the sample is positive. The method can be used for detecting a stimulant in competitive sports, is simple and feasible, can be used for rapidly detecting a great quantity of samples, and is flexible in detection and high in accuracy.
